| Viral myocarditis(Viral myocarditis, VMC) is the localized or diffuse lesions of myocardial caused by viral infection.In recent years, the incidence of viral myocarditis in children and young adults is rising year by year, and the disease has gradually become an important cause of death in children and adolescents.The serious and extensive myocardial tissue inflammation is heart failure which cause sharp decline of myocardial contractility, and it is important cause of death in patients. Currentlypathogenes-is of viral myocarditis complicated by heart failure is not clear, there is no effective treatment, to be studied specifically. Huangqi has significant effects of supplement Qi and strengthen the heart, a large number of reports indicate that Huangqi could treat viral myocarditis complicated by heart failure, but the drug active ingredient and mechanism of action is unknown.Objective:This study established a mouse model with viral myocarditis, observe the effect of ATF- effective ingredient of Huangqi to endoplasmic reticulum stress and mice with viral myocarditis ventricular myocytes L-type calcium current, L-type calcium channel expression。Methods: Adopting RT-PCR, Western blotting and other experiments to reserch the effects of to clear the treatment of viral myocarditis complicated by heart failure. Results:(1)Total flavonoids of Astragalus can reduce ventricular myocytes calcium channel opening time, increase closing time, reducing the open pro-bability of the mouse with viral myocarditis to inhibit calcium currents.(2) Total flavonoids of Astragaluscan reduce ventricular myocytes LCCsɑ1 subunit m RNA expression levels of the mouse with viral myocarditis to inhibit the overload of intracellular calcium.(3) Total flavonoids inhibit viral myocarditis mouse ventricular myocytes endoplasmic reticulum chaperones protein GRP78 expression.Conclusion: Total flavonoids of Astragalus improves hemodynamics in mice with viral myocarditis by the reasons as mentioned above. |
