Study On Micro Inflammatory State In The Optimization Of Dosage Form And Its Optimal Dosage Effect Of Yishen Jiangzhuo Prescription On Chronic Renal Failure Rats

Aim:Through observing the different curative effect on CRF rats models by giving different formulations of Yishen Jiangzhuo Decoction and influence of the micro-inflammatory state of CRF rats by giving the optimal formulations to discuss the mechanism about the treatment of Yishen Jiangzhuo Decoction for chronic renal failure.Methods:We take the method of 5/6 nephrectomy to copy animal models.84 SPF adult male SD rats,7 weeks age,weights vary from 180g to 200g,adaptively bred for 1 week,divided randomly into the blank group, sham operation group, model group, control group, each group has 12 rats, Yishen Jiangzhuo Decoction group (treatment group)36 rats.The Yishen Jiangzhuo Decoction group then divided randomly into decoction group, capsule group, granule group,each group has 12 rats.The rats of the model group、 the control group and the treatment group were taken operations of 5/6 nephrectomy; the rats of sham operation group were taken operations of stripping double renal envelope; we didn’t do any processing to the rats of the blank group.4 weeks after surgery,gave different treatments for 8weeks.At the fifty-seventh day collected urine and collected blood from abdominal aorta to isolate the serum,measured 24 hours urinary protein quantification (biuret method), blood routine and renal function,ELISA method for determined the content of the micro inflammation index CRP, TNF alpha, the content of IL - 6 and the content of ALB.Observed the pathological changes of renal tissue by light microscopy.Results:1. The RBC count and HB content of the model group significantly reduced,the serum Scr、 the serum BUN and the 24 hours urinary protein quantification significantly increased (p <0.05),the content of CRP、TNF-αIL-6 significantly increased (p<0.05),the content of ALB significantly reduced (p<0.05),Renal interstitial fibroblasts hyperplasia is active;2. The RBC count and HB content of the decoction group、capsule group、granule group and the control group increased,the serum Scr、the serum BUN and the 24 hours urinary protein quantification reduced (p<0.05),the proliferation and activation of the renal interstitial fibroblasts are suppressed,and the curative effect of the decoction group is better than the capsule group and the granule group Cp<0.05);3. The Yishen Jiangzhuo Decoction groups and the control group can reduce the serum CRP、TNF-a、IL-6 and increase the content of ALB (p<0.05),and the curative effect of the Yishen Jiangzhuo Decoction groups are better than the control group (p<0.05)Conclusion:1. The curative effect of Yishen Jiangzhuo Decoction of the decoction group is better than the capsule group and the granule group for the chronic renal failure;2. The Yishen Jiangzhuo Decoction can reduce the serum CRP、TNF-αIL-6 and increase the content of ALB,alleviate the development of chronic renal failure;3. The Yishen Jiangzhuo Decoction can alleviate the development of chronic renal failure,and can be an effect prescription of prophylaxis and treatment for chronic renal failure.
