| Objective: Ulcerative colitis(ulcerative colitis, UC) was a kind of unexplained chronic nonspecific inflammatory bowel disease, with the characteristic of various severity, hard to be cured and easy to relapse. It was regarded as one of the modern intractable diseases by WHO. The experiment by observing the change of Huazhuojieduyukui prescription in general situation of ulcerative colitis rats, disease activity index(DAI) scores, Colon histopathology, interleukin-8(IL-8) and Cyclooxygenase-2(COX-2), to explore the mechanism of Huazhuojieduyukui prescription for the treatment of ulcerative colitis.Methods: 60 male Wistar rats weighting between 250 to 300 g were daptive fed for one week. After being encoded, they were randomly divided into 6 groups, in which one group is 10, including the normal group(N), model group(M), the MS group(MS), Huazhuojieduyukui prescription high-dose group(HD), middle-dose group(MD), low-dose group(LD). Except the Normal group, other groups were built for UC animal model by using TNBS/ethanol. Since the first four days after modeling is completed, N group was normally fed, M group was given normal saline with 4ml/kg, the remaining four groups were respectively given drug gavage therapy for 2 weeks. During this period, observing the change in general situation of ulcerative colitis rats, DAI scores. After the treatment, killing rats and taking the blood to detect the content change of IL-8, dissecting colon tissue to make pathological observation and measuring the expression of COX-2, making statistical analysis of the resulting data in the end.Results:1 The general stateN group of rats were in good condition during the whole process, while others were depressed, unresponsive, reduced food intake, weight loss, messy fur and dark yellow light, dilute or purulent bloody stool.After the treatment, the spirit, intake, weight and the shape of feces in the groups of MS, HD, MD, LD groups were fairly improved. In all these groups, the most obvious improvement was HD group, the second were MS, MD groups and no significant difference between the two groups. Slightly less than other groups was LD group.2 DAI scoreMS, HD, MD and LD groups were all lower than M group, the difference was statistically significant(P<0.05); HD group was lower than MS, MD and LD groups, the difference was statistically significant(P<0.05); MS and MD groups were lower than LD group, the difference was statistically significant(P<0.05), however, the difference of MS and MD groups were no statistically significant.3 Pathology observationObserved under the microscope, the mucosal epithelium of N group looks smooth and full, the glands were clear and no other abnormalities; The number of mucosal ulcer in the M group was the most, the gland was unclear and badly damaged, the colon tissue was obvious congestion and edema, also there were a lot of inflammatory cells; MS, HD, MD and LD groups were better than M group under the microscope. The best was HD group, it was the closest to the N group. Mucous membrane was relatively smooth and there were the most new glands, the ulcer was superficial and in a healing period, the congestion, edema and inflammatory cells infiltration were not obvious; There is no significant difference between MD and MS group. In the two groups, there was a small amount of superficial ulcer, inflammatory cells infiltration and the mucous membrane’s congestion edema was mild, all these were less than HD group; In the LD group, the number of mucosal ulcer were slightly increased, the gland were slightly damaged and lack of new gland, inflammatory cells were increased and congestion, edema were visible.4 Biochemical testsIL-8: Compared with N group, other groups were increased to different extent in the content of IL-8. The M, MS, MG and LG group compared with N, there was statistically significant difference(P<0.05); HG group compared with N, there was no statistically significant difference(P>0.05); Compared with M group, the treatment groups was lower than M groups, there was statistically significant difference(P<0.05); Compared between treatment groups, HG group was lower than those of MD, MS and LG group, the difference was statistically significant(P<0.05); MS and MD group compared with LD group, the difference was statistically significant(P < 0.05); MD group compared with MS, there was no statistically significant difference(P > 0.05).COX-2: Compared with N group, other groups were increased to different extent in the content of COX-2. The M, MS, MG and LG group compared with N, there was statistically significant difference(P<0.05); HG group compared with N, there was no statistically significant difference(P>0.05); Compared with M group, the treatment groups was lower than M groups, there was statistically significant difference(P<0.05); Compared between treatment groups, HG group was lower than those of MD, MS and LG group, the difference was statistically significant(P<0.05); MS group compared with MD, LD group, the difference was statistically significant(P<0.05); MD group compared with LD, the difference was statistically significant(P<0.05).Conclusion:1 Huazhuojieduyukui prescription can significantly improve the general situation of ulcerative colitis rats and the colon tissue injury, decrease the DAI scores. There exist a certain dose-effect relationship that high-dose group is better than middle-dose and low-dose group.2 The mechanism of Huazhuojieduyukui prescription can treat UC may by decreasing the content of IL-8 and the expression of COX-2. By inhibiting the inflammatory response, reducing the inflammatory cell infiltration, blocking cell damage, it can effectively treat UC. |
