Purposes: To date,many interventional therapy researches have been carried out,but Restenosis after PCI is the key issues that will affect the long-term efficacy of the therapy. Although simvastatin could reduce the restenosis rate, but the effects should by improved and new drug should be found. The vascular intimal hyperplasia is the important characteristic of coronary atherosclerotic and vascular interventional operation. The mechanism of the vascular intimal hyperplasia contains endothelial injury and dysfunction, inflammatory reaction, platelet aggregation and vascular smooth muscle cells(VSMCs) migration et.al. The research results showed shenxiong glucose could affect hemorrheology, regulate lipid metabolism and reduce inflammatory reaction. The experiment established the test model of New Zealand rabbits’ with balloon injury and high fat-diet,the objective of this experiment is to observe the effects of Shenxiong glucose injection and simvastatin on carotid neointimal hyperplasia,and explore the possible mechanism. We hope to find new evidence of drug therapy to prevent the vascular restenosis after PCI.Methods: 4-month-old healthy male New Zealand rabbits were randomly divided into five groups:1 control group(C),which received high-fat diet;2 operation group(O),which received high-fat diet and abdominal aortic balloon endothelial injury surgery;3 simvastatin group(S), which received simvastatin 2.2mg/kg×d on the basis of operation group;4 shenxiong glucose group(S),which received shenxiong glucose injection 5ml/kg×d on the basis of operation group;5 combined treatment group(shenxiong glucose combined with simvastatin)(S+X),which received both shenxiong glucose and simvastatin the basis of operation group.Two weeks after operation and drug intervention, arteria carotid and blood were collected. Observation: 1) was used to detect the level of blood lipid; 2) H-E staining under the light microscope to observe the vascular intima; 3) immune group of determination of vascular intima and media expression of PCNA and MMP-9; 4) Western blot method for determination content of artery of GRP78 in. From four above aspects,we try to determine the intimal hyperplasia.Results:1 After 2 weeks of high-fat diet, the carotid artery intima hyperplasia was obvious, and the pathological staining showed intimal hyperplasia.2 There was no obvious intimal hyperplasia in the control(C) group, and the intimal hyperplasia was seen in the operation(O) group(P < 0.05).3 Shenxiong glucose(S)group, the vascular intima hyperplasia was significantly lower than that the surgery group and simvastatin group(P < 0.05).Shenxiong glucose + simvastatin(S + X) endometrium hyperplasia is lower than Shenxiong glucose(S) group, but no statistical difference.4 The control(C) group and operation(O) group between lipid levels are not significantly different, the lipid content of the simvastatin(X) group was significantly lower than the Shenxiong glucose(S) group and control(C) group and operation(O) group(P < 0.05). Shenxiong glucose group ’s(S) lipid content was lower than the controls(Group C) and operation group(O), but the difference between was of no statistical difference(P>0.05). Shenxiong glucose + simvastatin(S+X) group was lower than that of simvastatin(X) group, but without statistical significance.5 Under light microscope, PCNA, MMP9 immune group of positive cells were brown, control group(C) positive cells less, and operation group(O) and simvastatin group(X), Shenxiong glucose group(S) is visible vascular intima and media have a large number of positive cells. Compared to positive cells,Shenxiong glucose(S) group and operation group(O) were significantly reduced(P<0.05). Shenxiong glucose(S) group of MMP9 PCNA expression was lower than that of simvastatin(X) group, but without statistical significance. MMP9 PCNA expression of the Shenxiong glucose + simvastatin group(S+X) was lower than that of Shenxiong glucose(S) group, but without statistical significance.6 Shenxiong glucose(S) group of GRP78 expression level was significantly lower than that in operation(O) group and simvastatin(X) group(P< 0.05), and Shenxiong glucose + simvastatin(S + X) group expression was lower than that of Shenxiong glucose(S) group, but without statistical significance.Conclusion:1 High-fat diet and aortic balloon endothelial injury caused intima hyperplasia of arteries.2 The Shenxiong glucose can effectively reduce neointimal hyperplasia after balloon injury.The possible mechanism includes inhibited cell proliferation, hinder the smooth muscle cell migration and suppression of the ERS.
