| Objective To explored the importance of the miRNA-208a plasma concentrations in early diagnosis of acute myocardial infarction(AMI).Methods Fifty patients with chest distress and pain were enrolled,who were admitted to the People’s Hospital of Guangxi Zhuang Autonomous Region from December 2013 to December 2014.These patients were divided into AMI group (24pts) and UAP group (26pts) according to the clinical diagnostic standard and the coronary angiograhic results. In addition,10 healthy adults without cardiovascular diseases were enrolled as control group.The plasma concentrations of miRNA-208a were detected by quantitative real time polymerase chain reaction (RT-PCR) and the plasma concentrations of hs-cTnI and CK-MB were detected by enzyme linked fluorescent analysis methods(ELISA) in UAP group,healthy control group and at 3h,6h,12h,24h after the onset of the chest pain for AMI.The results were analyzed with statistical methods.Results (1) Plasma miRNA-208a level of the AMI group was obviously higher than that of UAP and the control groups (p<0.01). (2) The plasma miRNA-208a level was significantly increased at 3h after the onset of the chest pain for AMI,and reached its peak at 6h after the onset of the chest pain for AMI,then decreased to the normal level nearly at 24h after AMI. (3) ROC curve analysis revealed an area under the curve of 0.936 (p<0.01) for miRNA-208a, 0.891 (p<0.01) for hs-cTnI, and 0.910(p<0.01) for CK-MB. miRNA-208a was highly sensitive and specific for AMI patients within 3-6h after the onset of symptoms. (4)The miRNA-208a was positively correlated to the plasma hs-cTnI (r=0.20555,p<0.05) and CK-MB(r=0.28055,p<0.01).The miRNA-208a levels was not affected by age,gender,blood pressure, diabetes mellitus, smoking history,,Tcho, LDL, TG, CRP, kidney function.Conclusion The elevated cardiac-specific miRNA-208a in plasma may be a potential and novel biomarkers for early diagnosis of AMI. |
