Currently, study on mycotoxins in traditional Chinese medicine is in the transition stage from detecting a single species of mycotoxins to quantitatively analyzing a variety of mycotoxins. Since laws and regulations of mycotoxin for TCMs in our country is not as strict as that for foods, drug manufacturers lack of attention to this field which seriously restricts the development of the modernization and internationalization of TCM. Therefore, there are impressive needs to improve the quality control means and establish rapid quality evaluation standards for TCM production process.LC-MS is rapidly developed in recent years as a kind of detecting method, which has obvious advantage to LC in the determination of compounds of trace levels. Astragali radix is the dried root of leguminous plants Astragalus membranaceus (Fisch.)Bge.var.mongholicus (Bge.) Hsiao or Astragalus membranaceus (Fisch.)Bge., which is commonly used in clinical treatment with the ructions of diuresis, apocenosis, sore healing etc. Panax notoginseng is the dried root or rhizome of the panax quinquefolium panax notoginseng (Burk) F. H. Chen, which is native in the northeast of Yunnan, and some distributions of Guangxi and Sichuan.In this paper, Astragali radix, Panax notoginseng and their Chinese drugs preparations are used as the research object, based on LC-MS to establish the quantitative analysis method of 10 kinds of mycotoxins, which is of great significance to improve the quality of the related medicine. This study includes the following four aspects:(1) A rapid quantitative analysis method was developed to detect 4 kinds of aflatoxin (B1, B2, G1, G2), OTA, T-2 toxin, MPA, DON, ZAN and ZEN at the same time, using ultra high performance liquid tandem mass spectrometry technology in Astragali radix as the research object. Pretreatment method was optimized including experiments of chromatographic conditions, solvent extraction, solid phase extraction column and the solvent, the results showed that the linear relationships of ten kinds of fungus toxin were good, the correlation coefficients R2 were between 0.9920-0.9997, the recoveries were between 75.6%~117.0%, RSD were between 2.21%~14.3%. The method can be used to quantitatively analyze this ten kinds of mycotoxins in Astragali radix.(2) A rapid quantitative analysis method was developed to detect 4 kinds of aflatoxin (B1, B2, G1, G2), OTA, T-2 toxin, MPA, DON, ZAN and ZEN at the same time, using ultra high performance liquid tandem mass spectrometry technology in two kinds of Astragalus preparations as the research object. The results showed that the linear relationships of ten kinds of fungus toxin were good, the correlation coefficients R2 in astragalus injection were between 0.9951-0.9996, the recoveries were between 75.6%~117.0%; The correlation coefficients R2 in astragalus granule were between 0.9930~0.9999, the recoveries were between 83.5%-109.5%, RSD were between 7.5%~15.3%. The method can be used to quantitatively analyze this ten kinds of mycotoxins in these two astragalus preparations.(3) A rapid quantitative analysis method was developed to detect 4 kinds of aflatoxin (B1, B2, G1, G2), OTA, T-2 toxin, MPA, DON, ZAN and ZEN at the same time, using ultra high performance liquid tandem mass spectrometry technology in Panax notoginseng as the research object. The results showed that the linear relationships of ten kinds of fungus toxin were good, the correlation coefficients R2 were between 0.9981-1.0000, the recoveries were between 73.8%-105.6%, RSD were between 2.71%~6.75%. The method can be used to quantitatively analyze this ten kinds of mycotoxins in Panax notoginseng.(4) A rapid quantitative analysis method was developed to detect 4 kinds of aflatoxin (B1, B2, G1, G2), OTA, T-2 toxin, MPA, DON, ZAN and ZEN at the same time, using ultra high performance liquid tandem mass spectrometry technology inXuesaitong powder injection as the research object. The results showed that the linear relationships of ten kinds of fungus toxin were good, the correlation coefficients R2 were between 0.9988~0.9999, the recoveries were between 61.9%~106.6%, RSD were between 1.62%~6.13%. The method can be used to quantitatively analyse this ten kinds of mycotoxins in Xuesaitong powder injection.
