Objective:By observing the effect of platelet rich fibrin and Collagen membrane to repair the hard palate soft tissue defects in New Zealand rabbits, providing the reference for the PRF membrane repair soft tissue defects in oral cavity.Methods:Select 54 three-months old male New Zealand white rabbits which were randomly divided into 3 groups, respectively PRF group-, collagen membrane group and blank control group, with 18 in each group. In the front hard palate, respectively from the maxillary incisors, the back edge of the hard palate mucosa around 2mm ring with a diameter of 5mm tissue cutting drill preparing round full thickness defect of soft tissue. PRF membrane groups were implanted autologous PRF membrane in the defect, collagen membrane groups were implanted biofilm in the defect, blank control group without any treatment General observation was carried out on the wound and wound healing rate analysis after 3 days,7 days,14 days,21 days,28 days and 56 days.Three animals of each group were sacrificed that underwent surgery area drawn HE staining, Masson staining and CD31 immunohistochemical staining were observed in the surgical inflammation, collagen formation and angiogenesis. The data obtained using SPSS 17.0 analysis software for processing.Results:1. Morphological observation:In the early trauma, PRF group’s wounds reflect mild inflammatory reaction, collagen membrane group and blank control group’s inflammatory response were obvious.14 days after operation, PRF group’s wound epithelial were completely coveraged, no tissue depression, no scar formation. Collagen membrane group and the blank control group appeared different degree of organization in the process of healing sag and ultimately healing tissue scarring.2. Wound healing rate analysis:After three days, no obvious difference was found between different experimental group wound healing rate, does not have statistical significance (P> 0.05).7 days after surgery, wound healing rate of the PRF and collagen membrane group and blank control group differences between statistically significant (P< 0.05); PRF wound healing rate and collagen membrane group differences between statistically significant (P< 0.05), the size of the 7 days postoperative wound healing rate is:the PRF group> collagen membrane group> blank control group.3. Inflammatory pathology grade:3 and 7 days after surgery, there are differences between the three groups of inflammatory markers, with statistical significance (P< 0.05), comparing the two results are PRF group and the rest of the difference between the two groups, with statistical significance (P< 0.05); No difference between collagen membrane group and the blank control group, with statistical significance (P> 0.05). The 3 and 7 days after surgery, PRF inflammatory reaction is less than the collagen membrane group and blank control group. In postoperative days 14 and 21 days,28 days and 56 days, no difference between the three groups of inflammatory conditions, have no statistical significance (P> 0.05).4. Neovascularization:After 3 days,28 days and 56 days, the experimental group of CD31 mean optical density no significant difference was not statistically significant (P> 0.05).7 days and 14 days after surgery, PRF group CD31 mean optical density value is higher than the collagen membrane group and control group, the difference was statistically significant (P<0.05), collagen membrane CD31 group mean optical density value is higher than the control group The difference was statistically significant (P<0.05).21 days after surgery, PRF group CD31 mean optical density value is higher than the collagen membrane group and control group, the difference was statistically significant (P<0.05), collagen membrane CD31 group and control group, the mean optical density value was no significant difference, not statistically significant (P> 0.05).5. Average optical density of collagen fiber analysis:Three days after surgery, the experimental group of collagen fiber no difference between the average optical density value, do not have statistical significance (P> 0.05).7 days postoperatively, PRF set of collagen fiber average optical density value of the collagen membrane group and blank control group on the high side, with statistical significance (P< 0.05); Collagen membrane group compared with blank control group collagen fiber average optical density value is on the high side, statistically significant difference (P< 0.05).14 days postoperatively, PRF group, collagen membrane collagen fiber average optical density value higher than blank control group, with statistical significance (P< 0.05); PRF set of collagen fibers and the collagen membrane group no difference between the average optical density value, is not statistically significant (P> 0.05). After 21 days,28 days and 56 days, PRF group, collagen membrane collagen fiber average optical density value low compared with blank control group, difference has statistical significance (P< 0.05); PRF set of collagen fiber average optical density value is low in the collagen membrane group, difference has statistical significance (P< 0.05).Conclusion:1. PRF can promote faster oral soft tissue defects, without the presence of soft tissue healing phenomenon with depressed.2. PRF can alleviate the inflammatory reaction in the process of wound healing, especially in the early trauma (within one week after trauma), the effect is obvious.3. PRF can accelerate the process of wound newborn vascularization, promote early wound healing.4. PRF can promote wound collagen deposition in the early trauma, accelerating wound healing; In the middle and later periods of the trauma PRF can adjust the collagen metabolism, reduce scar formation, improve the quality of wound healing.
