Expression And Significance Of IL-12 In Classical Hodgkin Lymphoma And Diffuse Large B-cell Lymphoma

Object: To investigate the expression and clinical significance IL-12 in patients with classical Hodgkin lymphoma(CHL) and diffuse large B-cell lymphoma(DLBCL). Methods:(1) The expression of IL-12p40 and IL-12p70 protein were detected by Immunological Histological Chemistry(IHC) En Vision two-step method in 47 cases of CHL, 92 cases of DLBCL and 30 cases of reactive lymphoid hyperplasia(RLH).(2)The expression of IL-12p40 and IL-12p70 m RNA were detected by the In Situ Hybridization(ISH) technique in 47 cases of CHL, 50 cases of DLBCL and 30 cases of RLH. Results:(1) The positive rates of IL-12p40 and IL-12p70 protein were 53.19%(25/47), 70.21%(33/47) in CHL and 0.00%(0/92), 4.35%(4/92) in DLBCL, respectively. No expression of IL-12 protein was observed in RLH. The expression rates of IL-12p40 and IL-12p70 protein in CHL were significantly higher than those in DLBCL and RLH(P <0.05). There is no significant correlation between IL-12p40 and IL-12p70 protein with sex, age, bulky disease, B symptom, clinic stage, serum LDH level and international prognostic score(IPS). IL-12p40 protein was positively correlated with the expression of IL-12p70 protein in CHL.(2) The positive rates of IL-12p35 and IL-12p40 m RNA were 31.91%(15/47), 44.68%(21/47) in CHL. No expression of IL-12 m RNA was observed in DLBCL or RLH. The expression rates of IL-12p35 and IL-12p40 m RNA in CHL were significantly higher than those in DLBCL and RLH(P <0.05). There is no significant correlation between IL-12p35 m RNA and IL-12p40 m RNA with sex, age, bulky disease, B symptom, clinic stage, serum LDH level and international prognostic score(IPS). IL-12p35 m RNA was not correlated with the expression of IL-12p40 m RNA in CHL. Conclusion:(1)The high expression ofIL-12p40 and IL-12p70 protein in HRS cells might contribute to the development and genesis of micro-environment of CHL and provide new experimental basis for immunotherapy of CHL.(2)Detection of IL-12p35 and IL-12p40 is helpful for the diagnosis and differential diagnosis of CHL.
