| The illegal fortification of chemical medicines in natural dietary supplements is a growing concern in most country, especially for antihypertensive and hypoglycemic drugs. Prohibited chemical drugs were added into natural dietary ingredients for the great benefits. Addationally this kind of chemical drugs abused for a long time will result in serious consequences. So it was very necessary to develop effective detection method for detection of antihypertensive and hypoglycemic drugs.To date instrument analysis is the main detection method for antihypertensive and hypoglycemic drugs, for example HPLC, LC/MS etc. There were a lot of disadvantages for instrument analysis in rapid test, including the require of well trained operators, high cost, time-consuming, complex pretreatment. In this research, we aim to develop a rapid, sensitive and specific chemiluminescence immunoassay for determination of repaglinide, nateglinide and captopril.Immunogens were synthesized by coupling the thiol groups in captopril, carboxyl groups in repaglinde and nateglinide with carrier proteins. Polyclonal antibodies were produced, and the combinations of antibodies and coating antigens were screened to get the optimal combination. The sensitivities of captopril, repaglinide and nateglinide were 1110.92,40.05,72.33 ng/mL respectively in ELISA. These important factors of CLIA were optimized by the control variable method. And it was found that CLIA was better than ELISA significantly. The sensitivities and linear ranges were 193.54 ng/mL,10.16-3685.54 ng/mLã€28.58 ng/mL,1-180 ng/mL,4.68 ng/mL,0.1-100 ng/mL for captopril, repaglinide and nateglinide respectively. Compared with ELISA, the sensitivities were much higher and linear ranges were much wider.In summary, a kind of sensitive, rapid, special and effective immunoassay was established to detect the addition of antihypertensive and hypoglycemic drugs in foods, and suitable for high throughput screening. |
