Objective:1.Through the herbalogical study and many kinds of identification, to identify the quality distinction of Dendranthema morifolium(Ramat.) Tzvell’Gongju’ cv.nov. and its adulterant Dendranthema morifolium(Ramat.) Tzve ’Qiyueju’ cv.nov.2.To optimize the processing technique method of genuine medical herb Dendranthema morifolium(Ramat.) Tzvel’Gongju’ cv.nov.3.To establish the exclusive quality evaluation of genuine medical herb Dendranthema morifolium(Ramat.) Tzvel’Gongju’ cv.nov., which accomplished the excellent quality assessment of Dendranthema morifolium(Ramat.) Tzvel’Gongju’ cv.nov..Methods:1.Through the herbalogical study, the name, breeds, place of origin of Dendranthema morifolium(Ramat.) Tzvel’Gongju’ cv.nov. were discussed in this paper, which was to identify the origin and distinction with its adulterant Dendranthema morifolium(Ramat.)Tzvel ’Qiyueju’ cv.nov. Used traits micro, microscopic characteristics, TLC, GC-MS component analysis and other means of identification,which was to find the quality distinguish points of ’Gongju’ and ’Qiyueju’.2.Collected ’Gongju’ and ’Qiyueju’ samples from Huangshan city in Anhui province, the optimum picking period, drying sthyle, microwave drying conditions of those were discussed,which was to establish the efficient processing technique method. The method could maintain high quality fresh colors of ’Gongju’.3.According to the methods of description and the thin layer chromatography(TLC) in“Chrysanthemum morifolium Ramat.” in the Chinese Pharmacopoeia(I,2010 Edition),the description and TLC of ’Gongju’ and ’Qiyueju’ were obtained; According to technical requirements in the appendix in the Chinese Pharmacopoeia(I,2010 Edition).The extractives, moisture, total ash, ulfur dioxide residues, heavy metals(As, Cd, Hg,Pb,Cu), the content of aflatoxins were obtained; established the HPLC method fordeterminating simultaneously luteolin, galuteolin and chlorogenic acid, 3,5-O-dicaffeoylquinic acid mehtylester in ’Gongju’ and ’Qiyueju’, and compared the content distinction in ’Gongju’ and ’Qiyueju’; Used normal phase HPLC to study HPLC-fingerprint of ’Gongju’ and ’Qiyueju’ by using vitamin A as marker compound,which established HPLC-fingerprint method of ’Gongju’.Results:1. Through the herbalogical study, drawn: ’Gongju’ was original from Song, abundant from Ming and Qing, after being fame as a tribute to the palace, known as the " Gongju". And ’Qiyueju’ was come from the culturing of ’Gongju’, forming two flowering,because the first flowering in July, known as the ’Qiyueju’.Used traits micro,microscopic characteristics, TLC, GC-MS component analysis and other means of identification,to find distinguish points of ’Gongju’ and ’Qiyueju’ in description, TLC and GC-MS component analysis.2. Through the processing technique study of ’Gongju’ and ’Qiyueju’ from Huangshan city, established the efficient processing technique method that could maintain high quality fresh colors of ’Gongju’: ①the optimum picking period: ’Gongju’ was picked in November, the first harvest flowers were better, the third harvest flowers should not be used; ’Qiyueju’ in August was best, the first harvest flowers were better, the third harvest flowers should not be used. ②microwave filming and drying conditions:Chrysanthemum morifolium Ramat.was sealing film sealed 2 min for microwave filming on 80℃, the color of Chrysanthemum morifolium Ramat.was best, airing 2min.It was microwave drying 27 min on 55℃, airing again one night, secondary drying:heating 8 min on 55℃ after cooling to room temperature.3.Established the exclusive quality evaluation of genuine medical herb ’Gongju’ :By description found ’Gongju’ and ’Qiyueju’ had differences in capitulum size, half flourish size, number of tubular flowers, nature, air, taste aspects, and revised the description of ’Gongju’ in 2010 edition of "Chinese Pharmacopoeia".By TLC identification method, chlorogenic acid as reference substance andChrysanthemum morifolium Ramat. as contrast, found 5 batches of ’Gongju’ had significantly spots, 3 batches of ’Qiyueju’ had not significantly spots, which could distinguish obviously with ’Gongju’.Measured 21 batches of ’Gongju’:The water-soluble extractives content of22.70%-24.64%, ethanol-soluble extractives content of 19.99%-21.27%, moisture content of 8.42%-10.61%, total ash content of 4.91%-6.13% was obtained. 12 batches of ’Qiyueju’: The water-soluble extractives content of 14.92%-17.58%, ethanol-soluble extractives content of 11.56%-17.94%, moisture content of 8.53%-11.56%, total ash content of4.94%- 6.17% was obtained. The content of Pb、Cd、As、Hg、Cu of 21 batches of ’Gongju’ were 0.37、0.425、0.357、1.034、9.039mg/kg, respectively. The content of Pb,Cd,As,Hg,Cu of 12 batches of ’Qiyueju’ were 0.960、1.370、0.572、0、17.507mg/kg,respectively.The aflatoxins and ulfur dioxide residues were not detected in 21 batches of’Gongju’ and 12 batches of ’Qiyueju’.Established the HPLC method for determinating simultaneously chlorogenic acid,galuteolin, 3, 5-O-dicaffeoylquinic acid mehtylester and luteolin in ’Gongju’ and’Qiyueju’, found the content of chlorogenic acid and 3, 5-O-dicaffeoylquinic acid mehtylester in two kinds of Chrysanthemum morifolium Ramat. were approach, the content of galuteolin and luteolin had obviously difference. The content of galuteolin in’Gongju’ was higher than that of ’Qiyueju’, and the content of luteolin in ’Qiyueju’ was higher than that of ’Gongju’. Compared and analyzed the intrinsic quality of influence that the processing technology were used the 4 effective components of ’Gongju’ and’Qiyueju’, thus to select the efficient processing technique method that could maintain high quality fresh colors of ’Gongju’.Determined 7 mutual peaks on the chromatographic fingerprint, taked the vitamin A chromatographic peak as reference peak, established the normal phase HPLC-fingerprint method of ’Gongju’ and ’Qiyueju’, the similarities of 21 batchs of’Gongju’ were more than 0.95, the similarities of 12 batchs of ’Qiyueju’ were more than0.8, the similarities of two kinds of Chrysanthemum morifolium Ramat. were lower than0.7.Conclusion:The facts mentioned above illuminate that ’Gongju’ and its adulterant ’Qiyueju’ had obvious difference on the trait, TLC chromatography, GC-MS composition analysis, the content of total flavonoids. Established the quality evaluation system of genuine medicinal materials ’Gongju’. These methods are strong characteristics, accurate,reliable, good reproducibility and can be used for the standards and the quality estimation about the genuine medicinal materials ’Gongju’.
