| Research 1 Changes over time of CB1R in hippocampus of Status Epilepticus ratBackground and purpose:Epilepsy is one of the most common disease of neurology. Status Epilepticus (SE) is a kind of emergency disease, it is very common in clinical, and with a high mortality and morbidity. Benzodiazepines drugs are the first choice to control SE, but most clinical studies have shown that the efficient of Benzodiazepines drugs will decline if delay to give drugs to patients of SE. The endogenous cannabinoid may activate presynaptic CB1 receptors in a retrograde manner, resulting in a reduction of neurotransmitter release at various synapses. This report aimed to explore the changes over time of Cannabinoid Receptor 1 (CB1R) in hippocampus of SE and the role of CB1R in the generation of epilepsy, to looking for new targets for the treatment of epilepsy.Methods:Models of SE were induced by intraperitoneal injection of Kainic Acid (KA), SE were terminated 30 minutes later, then the changes of CB1R were detected by immunohistochemistry and Western-blot at 1hour,1day, 1week,1 month,2 months after the termination of Status Epilepticus. In contrast, statistical analysis of data from One-Way ANOVA, One-Sample T Test and Rank-Sum Test.Results:1. CB1R in CA1 area of hippocampus of SE rats was increased gradually in 1 week, and then decreased to normal after 1 week (KA vs. NS:5.5±1.1 vs.1.8±0.2,6.4±3.7 vs.3.1±0.7,16.9±10.4 vs.3.7±1.7,8.8±5.4 vs.6.9±4.0,3.2±1.0 vs.4.4±1.9). The IA of CB1R was insignificantly increased in 1week (p<0.05), and insignificantly deceased after 1 week (p>0.05).2. CB1R in CA3 area of hippocampus of SE rats was increased gradually in 1 week, and then decreased to normal after 1 week. And CB1R in the area of CA3 was the same trend with area of CA1. The IA of CB1R was significantly increased in 1week (p<0.05), and insignificantly deceased after 1 week (p>0.05).3. CB1R in DG area of hippocampus of SE rats was increased gradually in 1 week, and then decreased to normal after 1 week. And CBIR in the area of DG were the same trend with the area of CA1 and CA3. The IA of CB1R was significantly increased in lweek (p<0.05), and insignificantly deceased after 1 week (p>0.05). Conclusion:The change of CB1R in hippocampus of SE rats was increased for protection, and then return to normal, it shows that CB1R may involved in the seizures and seizure free.3. CB1R in DG area of hippocampus of SE rats was increased gradually in 1 week, and then decreased to normal after 1 week. And CBIR in the area of DG were the same trend with the area of CA1 and CA3. The IA of CB1R was significantly increased in lweek (p<0.05), and insignificantly deceased after 1 week (p>0.05). Orexins may activate presynaptic orexin receptor-1 (OX1R) or orexin receptor-2(OX2R), and then resulting in a reduction of neurotransmitter release at various synapses. OX1R and OX2R are highly expressed in hippocampal neurons. A report has shown that the concentration of Orexin-A is decline in patients with repeated seizure, it may hint that Orexin-A may have special association with epilepsy. Orexin-A is mainly act on OX1R, and the affinity of OX1R is ten times than OX2R.
