Objective: To study the effect of aprednisolon combined with methotrexate on rheumatoid arthritis synovial fibroblasts for Treg/Th17 immune balance. Methods: Separation, purification, culture synovial tissue of patients with rheumatoid arthritis synovial cell liked fibroblast. Then cell were treated with aprednisolon and methotrexate. FLS were incubated with different concentrations of FSN and MTX, namely FSN 1, 2, 3 groups;MTX 1, 2, 3 groups and the combination 1, 2, 3 groups. The medium without drugs was used as a control group. There was a total of 10 groups. FLS proliferation was detected by MTT method; IL-10, IL-17, Foxp3, ROR-γt were detected by real-time PCR method, IL-10, IL-17,IL-22 were detected by ELISA method. Results:(1): proliferation: FSN group, MTX group, combined group FLS proliferation OD value were significantly lower than that control group; especially the combined 2 group. The difference was statistically significant(P<0.05).(2) mRNA content: Compared with the control group, the IL-10 and Foxp3 mRNA expression of combined group 2 were increased,The IL-17 and ROR-γt mRNA expression of combined group 2 was decreased;The difference was statistically significant(P<0.05).(3)ELISA: Compared with control group, the IL-10 levels of combined group 2 were increased; The IL-17 and IL-22 levels of combined group 2 were significantly decreased; The difference was statistically significant(P<0.05). Conclusion: Aprednisolon combined with methotrexate can inhibit RA FLS proliferation, elevated IL-10、Foxp3 expression, reduced the IL-17、ROR-γt、IL-22 expression, adjust Treg/Th17 immune balance; effect of 0.1mg/ml FSN +0.1mg/ml MTX is the most obvious.
