Objective: To investigate the effect of freezing factor on the survival rate and developmental potential of mouse oocyte during vitrification. Methods:â‘  according to the different equilibrium time were divided into three groups: oocytes were divided into fresh group, the equilibrium time of 5, 10 and 20 min group(I, II, III group), observed I, II, III egg mother cell survival rate, zona rupture rate, rupture of the plasma membrane, perivitelline space broadening rate; for the post thaw survival of oocytes were fertilized, cultivate and fresh group to investigate the developmental potential.â‘¡according to the different carriers: open carrier(OPS) and closed(CPS) carrier were cryopreserved with cumulus cells(COC) or stripping removal of cumulus cells(OD) of mouse cumulus oocyte complexes, recovery after respectively in vitro fertilization-embryo culture were observed and the survival rate and growth potential.Results: after vitrification freezing saves the oocyte survival rate between I and II group was statistically significant(81.9%vs57.3%, X2=7.13, P < 0.05); between II and III group have statistical significance(57.3%vs25%, X2=8.22, P < 0.05); between I and III group have significant statistically(81.9%vs25%, X2=17.92, P < 0.01).The rupture rate, the rupture rate of the membrane and the wide rate of the egg circumferential gap were not statistically significant due to the number of samples.Fresh and frozen group between fertilization rate was statistically significant(71.6%vs36%, X2=13.92, P0.01) cleavage rate, high-quality embryo rate differences were not statistically significant(97.6%vs94.4%, X2=1.82, P > 0.05; 95.2%vs88.2%, X2=2.16, P > 0.05). Survival rate in group II OPS is slightly higher than that in the CPS group, no significant difference(68%vs65.3%, X2=1.32, P > 0.05).Ops group COC group recovery rate was significantly higher than that of the dissolved oxygen(do) group, a statistically significant difference(78.6%vs57.4%, X 2=11.82, P < 0.05), and COC group and dissolved oxygen(do) group of fertilization rate and blastocyst rate compared to, there was no significant difference(P > 0.05).In the CPS group COC group recovery rate(75.4%) was significantly higher than that of dissolved oxygen(do)(55.1%) group, a statistically significant difference(75.4%vs55.1%, X2=11.98, P < 0.05), and two groups of fertilization rate and blastocyst formation rate had no significant difference, and OPS group whether package granule cells frozen storage, consistent with the conclusion.Conclusion: â‘ the appropriate pre balance time can avoid the freezing injury of mouse oocyte.Equilibrium time for oocytes of 5min groupThe highest survival rate of cells.â‘¡frozen thawed the survival of fertilized eggs after fertilization rate decreased, but after fertilization, the embryo developmental potential seemed not to be much of an impactâ‘¢the closed vector can effectively avoid the potential pollution problems, but different carriers to egg survival rates were not significantly influenced.â‘£The cumulus cells and oocytes frozen in glass It can play a protective role in the cell, and have some improvement to the development of the embryo.
