| Objective:the effect of Tianxiangdan Granule on the expression of MMP-9 and TIMP-1 in atherosclerotic HuiZhuoTanzhu syndrome of blood lipid and plaques in ApoE-/-mice. Methods:ApoE-/-mice were blank control group fed normal diet, model group ApoE-/-mice with high fat feed composite dry and cold environment and fed for 12 weeks to construct atherosclerosis HuiZhuoTanzhu syndrome model, the rats were randomly divided into Tianxiangdan group, atorvastatin group, model group.12 weeks later, the eyeball blood was collected to determine the content of lipid, aorta arch electron microscopic observation on pathological changes. The aortic arch portion HE of the morphological observation and image analysis for determination of dyeing, plaque area and lumen area ratio. The expression of immunohistochemical method for MMP-9 and TIMP-1 detection of plaque tissue. Results:after 12 weeks, groups of ApoE-/-mice there was no significant difference between water quantity. Compared with the blank control group, model group reduced food intake, Tianxiangdan group, atorvastatin group compared with the model group, no statistical significance. Compared with the control group, the final body weight, growth rate of body weight, serum TC, LDL content, plaque area/lumen area and plaque MMP-9 ratio increased in mice of model group, the TIMP-1 ratio decreased. Compared with model group, Tianxiangdan group mice at the end of body weight, body weight growth rate decreased, TC and LDL content, plaque area/lumen area and plaque MMP-9 ratio decreased, the ratio of TIMP-1 was increased (P<0.01), TG, no statistical significance of HDL content. Conclusion:Tianxiangdan granule can effectively reduce atherosclerosis HuiZhuoTanzhu syndrome of ApoE-/-mouse TC and LDL content, the ratio of plaque area/lumen area, decrease the expression level of MMP-9 in plaque, increased levels of TIMP-1 expression, thereby inhibiting collagen decomposition, has the stability effect on atherosclerosis plaque HuiZhuoTanzhu syndrome. |
