Object:To observe the effect of different acupuncture methods on wound healing related cells of model rats (Fibroblasts) and growth factor(FGF), To explore the mechanism of electroacupuncture in different wound healing, for the clinical treatment of difficult healing of skin wound and provide new ideas.Methods:To select 80 Wistar rats of clean grade, Male female half and half.The rats were divided into positive group, negative group, a surrounding needling group and blank group,20 rats in each group, Each group was divided into 1 days,3 days,7 days,10 days,and 15 days.4 rats at each time point. After the model of four groups were fixed, fixed method used in wood, Acupuncture needle acupuncture with the 0.5 inch Hua Tuo, at a depth of 0.2 inches, The positive group was tied in the center of the wound a needle, the center of the wound left side opening 0.5cm binding stitch, needle direction towards the direction of the needle wound, even the middle electromagnetic stimulator cathode, the left needle is connected to the negative; Negative group and positive group tied the same, only the middle pin is connected to the negative, the left needle is connected with the anode; Surrounding needling group the first needle in the center of the wound left 0.5cm, second pin 0.5cm in the center of the wound open right side, two needles are directed to the wound left side connected to the positive direction of needle, connecting the cathode right; The blank control group with normal fixed, but not by Electroacupuncture stimulation. The output current of electroacupuncture is 0.5 A, the frequency is 0.5Hz, the retaining time of 30 min,1 times a day. Each group were harvested at each time point, HE staining and immunohistochemical procedures were compared.Results:In the treatment of the first day of positive group, negative group, confining puncture group and blank group four groups around the wound fibroblasts were compared, there was no significant difference had no significance (P<0.05).Then negative group, positive group and blank group and positive group, Round needling group respectively, and compared with the control group no significant difference,7 days,10 days,15 days group three groups of time cut-off point of comparison showed very significant difference (P<0.01).At the 10 day of cutting, the difference between the stab group and the blank group was also visible(P<0.05).At the 15 day, the difference between the stab group and the blank group was significantly different (P<0.01).Predictably with increase of electroacupuncture treatment, the therapeutic effect of electroacupuncture highlighted gradually, especially from the third day began to negative group began to appear,From the seventh day to the set of positive role gradually, from the tenth day time section surrounding needling group have gradually emerged in the effect is obviously superior to the control group,Thus, electro acupuncture can promote the wound healing obviously.Conclusions:(1) Electro acupuncture therapy can significantly promote wound healing, shorten the wound healing time.(2)This experiment can prove that the anode in the wound in the middle of the electro acupuncture therapy by accelerating fibroblast formation rapidly promote rat wound healing, which may be one of the mechanisms of the effect of rat skin wound electroacupuncture in treating.
