| Objective:To evaluate the cytotoxicity of hydroxyapatite surface constructed by piezoelectric ceramic coating in vitro.Methods:The 3rd generation beagle bone marrow mesenchymal stem cells(BMSCs) were seeded on hydroxyapatite surface const-ructed by piezoelectric ceramic coating speciments and hydroxyapatite speciments,respectively. After 5 days,the cell adhesion was detected by scanning electron micro-scopy.The 3rd generation beagle bone marrow mesenchymal stem cells(BMSCs) were also co-cultured with hydroxyapatite surface constructed by piezoelectric ceramic coating speciments extract,hydroxyapatite speciments extract,low-glucose Duibecco’s modified Eagle’s medium containing 5% dimethylsulfoxide and 15% fetal bovine serum,and low-glucose Duibecco’s modified Eagle’s medium containing 15% fetal b-ovine serum,respectively.The cytotoxicity was tested by Cell Counting Kit-8 assay at days1,3,5after co-coulture.Results:Bone marrow mesenchymal stem cells on the s-urface of hydroxyapatite surface constructed by piezoelectric ceramic coating speciments and hydroxyapatite speciments grew proliferatively and presented with mulyil-ayergrowth.The connection between cells and pseudopodia was very close.Cell Cou-nting Kit-8 assay showed that the cells cultured in the extracts of hydroxyapatite surface constructed by piezoelectric ceramic coating and hydroxyapatite speciments proliferated more than 80%.Conclusion:The results above indicate that the two kind-s of materials both have good cytocompatibility and the toxicity was grade I that meant no cytotoxity. |
