Objective: To investigate the absorption of alliin, Research the solubility of alliin, using in vitro, in vivo, Caco-2 cells are three ways to study its absorption. Methods: 1. Using Fiber-optic sensing intrinsic dissolution tester(FOIDT) determination of alliin intrinsic dissolution rate, its research in four kinds of solvent in the dissolution behavior and intrinsic dissolution rate(IDR). 2. Using Optical Fiber sensing process analysis system combine with Everted Gut Sac model, real-time and in-situ determination the concentration of alliin in the rabbit intestine, study the absorption in different bowels and different concentrations. 3. Using Single Pass Intestinal Perfusion model of alliin in rat intestinal absorption, formulated with different concentrations of alliin respectively, determinate the effective permeability coefficient(Peff) and absorption rate constant(Ka) of intestinal in rats. 4. Using Caco-2 cell model, cell toxicity test(MTT) to determine the cell to the safe concentration of alliin, measured the apparent permeability coefficient(Papp) and intake of alliin in Caco-2 cells. Results: 1. Alliin materials in different solvents in completely dissolved within 2 min, the four kinds of solvent dissolution behavior similar, and both have good dissolution properties. Alliin in water, artificial gastric juice, acetate buffers, artificial intestinal juice four kinds of solvent the intrinsic dissolution rate is(43.48, 47.34, 45.06, 41.22 mg/min/cm2) respectively. 2. The absorption capacity of alliin in different rabbits bowel: jejunum> ileum > colon > duodenum; the absorption number An values of different concentrations in jejunum and ileum were greater than 1.15, duodenum and colon were less than 1.15. 3. Alliin in rat jejunum and ileum the Peff values were greater than 2×10-4 cm/s in these two belong to the high permeability of the intestines, the Peff values of colon and duodenum greater than 0.03×10-4cm/s but less than 2×10-4 cm/s, does not belong to the low permeability. 4. The Papp values of alliin in Caco-2 cells are greater than 1 ×10-6cm/s, suggesting that the alliin has good absorption in the Caco-2 cells. Intake with concentration and time dependent manner, PCaco-2(Bâ†'A) /PCaco-2(Aâ†'B) <1.5, suggesting that alliin in Caco-2 cells may be passive diffusion transport. Conclusion: 1.The fiber-optic in situ intrinsic dissolution test system can measure the intrinsic dissolution of alliin. 2. Using Fiber Optic sensing system determination of the concentration of alliin in the intestine in real-time, thus study its absorption in different intestines bowel, the results showed permeability of alliin in rabbit intestinal except the duodenum other is very good. 3. Alliin permeability in rat has good absorption properties. 4. Alliin has good permeability in Caco-2 cells with good absorption properties, the transport mechanism may be passive diffusion.
