| Objective:This study proposed the q RT-PCR, Western blot and Immunohistochemistry methods to test the expression of ARHGEF-7 in hepatocellular carcinoma tissues, adjacent to carcinoma and normal liver tissues in m RNA and protein level, and analyzed that the relationship between the expression of ARHGEF-7 and the clinical pathological features in hepatocellular carcinoma, to explore the clinical significance of ARHGEF-7 in hepatocellular carcinoma. In order to lay the foundation for further research of ARHGEF-7 in hepatocellular carcinoma.Methods:1.The q RT-PCR was used to detect the expression levels of ARHGEF-7 m RNA in 30 cases of HCC tissues, 30 cases of liver tissues adjacent to carcinoma and 5 cases of normal liver tissues, and to analyze the relationship between ARHGEF-7 and clinical pathological features of HCC2.Western blot was used to detect the expression levels of ARHGEF-7 protein in 30 cases of HCC tissues, 30 cases of liver tissues adjacent to carcinoma and 5 cases of normal liver tissues, and to analyze the relationship between ARHGEF-7 and clinical pathological features of HCC.3.Immunohistochemistry method was used to detect the expression intensity of ARHGEF-7 protein in 80 cases of HCC paraffin sections, and to locate the expression part of ARHGEF-7 in the cell, and to analyze the relationship between the expression of ARHGEF-7 and the clinical pathological features of HCC.Results: 1.The results of q RT-PCR showed that ARHGEF-7 m RNA expressed in 30 cases of HCC tissues, 30 cases of liver tissues adjacent to carcinoma and 5 cases of normal liver tissues and any comparison between the two groups, the difference was statistically significant(P<0.05). ARHGEF-7 m RNA expression in the three groups: HCC tissuesï¹¥liver tissues adjacent to carcinomaï¹¥normal liver tissues.The expression levels of ARHGEF-7 m RNA in multiple tumor nodules, Edmondson-Steiner grade â…¢-â…£stage and venous invasion tissues was significantly higher than those of HCC organizations which were single nodule, Edmondson-Steiner gradeâ… -â…¡stage and no vein invasion tissues.The expression levels of ARHGEF-7 m RNA in HCC tissues had no relationship with gender, age, the presence of cirrhosis of the liver and presence of plasma membrane and other clinical pathological features(P > 0.05).2.The results of Western blot showed that ARHGEF-7 protein expressed in 30 cases of HCC tissues, 30 cases of liver tissues adjacent to carcinoma and 5 cases of normal liver tissues. In HCC tissues, the expression levels of ARHGEF-7 protein were obviously higher than those of liver tissues adjacent to carcinoma and normal liver tissues. Similar to q RT-PCR, ARHGEF-7 protein expression in the three groups: HCC tissuesï¹¥liver tissues adjacent to carcinomaï¹¥normal liver tissues.The expression levels of ARHGEF-7 protein in multiple tumor nodules, Edmondson-Steiner grade â…¢- â…£ stage and venous invasion tissues was significantly higher than those of HCC organizations which were single nodule, Edmondson-Steiner grade â… - â…¡ stage and no vein invasion tissues.The expression levels of ARHGEF-7 protein in HCC tissues had no relationship with gender, age, the presence of cirrhosis of the liver and presence of plasma membrane and other clinical pathological features(P > 0.05). 3.Immunohistochemistry showed that in 80 cases of HCC tissues, ARHGEF-7 protein positively expressed in 67 cases and the expression intensity score 3+,2+and 1+in 19, 28 and 20 cases respectively.The other 13 cases were negative expression. The ARHGEF-7 protein were mainly located in cytoplasm.The expression intensity of ARHGEF-7 protein in multiple tumor nodules, Edmondson-Steiner grade â…¢-â…£stage and venous invasion tissues was significantly higher than those of HCC organizations which were single nodule, Edmondson-Steiner gradeâ… -â…¡stage and no vein invasion tissues.The expression intensity of ARHGEF-7 protein in HCC tissues had no relationship with gender, age, the presence of cirrhosis of the liver and presence of plasma membrane and other clinical pathological features(P > 0.05).Conclusion : 1.The expression levels of ARHGEF-7 m RNA and protein in HCC tissues were significantly higher than liver tissues adjacent to carcinoma and normal liver tissues2.The expression levels of ARHGEF-7 in HCC tissues had closely relationship with multiple tumor nodules, Edmondson-Steiner grade â…¢-â…£ stage and venous invasion. |
