Therapeutic Efficacy Of An Oncolytic Adenovirus Ad-HP In Gastric Carcinoma

Objective:To research the inhibitory effect of a expressing HN protein oncolytic adenovirus Ad-HP on human gastric cancer in vitro and in vivo.Methods:The SGC7901 cells inhibition was examined by MTS method; Suppression methods of Ad-HP on human gastric cancer cells were detected using by AO/EB staining, DAPI staining and Annexin V; Inhibition pathways of Ad-HP on human gastric cancer cells were detected by Western blotting; Inhibiting effection of Ad-HP on human gastric cancer cells was detected by establishing subcutaneous transplantation tumors.Results:Ad-HP can effectively inhibit the proliferation of SGC7901 cells; Ad-HP may through activation of caspase enzymes induce apoptosis in SGC7901 cells; Ad-HP can significantly extend the model animals’survival time, and effectively inhibited tumor growth; Ad-HP activated NK cells, and stimulated the production of IL-2, IL-4, IL-10 and IFN-y.Conclusion:Methodology validation of MTS, AO/EB staining, DAPI staining, Western-blot which were used for detecting inhibitory effect of Ad-HP on human gastric cancer cells was establish, and have a good inhibitory effect of Ad-HP on human gastric cancer cells. This article provides a foundation for the research the inhibitory effect of other oncolytic adenovirus on other tumor cells in the laboratory.
