| ObjectiveInvestigating the expression of Zinc-finger protein X-linked in the tissue and adjacent normal tissue of colorectal cancer, analyzing its correlation to the clinicopathologic features and exploring the factors influencing the prognosis of colorectal cancer,in order to provide potential target for the diagnosis, treatment and prognosis of colorectal cancer.MethodsA total of 35 pairs of colorectal cancer cases who underwent surgical resection in general surgery department of Shanghai Sixth People’s Hospital from June 2013 to January 2014 were collected, every pair of colorectal cancer case had received colorectal cancer tissue and adjacent normal tissue for Real time PCR and Western blot experiments.140 pairs of colorectal cancer cases(colorectal cancer tissue and adjacent normal tissue) were selected from september 2002 to august 2007, each pair had complete clinicopathological and follow-up data. Use colorectal cancer cases as test group, the adjacent normal tissue as control group. Detected the expression level of test group and control group through and immunohistochemistry method. Colorectal cancer cases were divided into high expression group and low expression group according Zfx expression level, its relationship with clinicopathological features were also investigated.Overall survival and disease-free survival analysis with Kaplan-Meier method plotted survival curves,the difference was significant by log-rank test for comparison. Multivariate analysis using multivariate Cox proportional hazard model to select independent prognostic factors.Results1. The expression levels of Zfx m RNA in colorectal cancer tissues and adjacent tissues measured by PCR and Western blot methods were 1.13 ± 0.42 vs 0.23 ± 0.12(P <0.001)and 0.97 ± 0.37 vs 0.61±0.18(P=0.005).The high expression number of cases detected by immunohistochemistry in colorectal cancer tissues and adjacent tissues were 76/140 vs 17/140(P<0.001).2. The expression level of Zfx was significantly associated with tumor differentiation, lymph node metastasis and TNM stage(P=0.001,P=0.017,P=0.002). However, there was no correlation between Zfx expression and other clinicopathological parameters, including gender,age,tumor location and tumor size(P=0.235,P=0.491,P=0.611,P=0.089).3. Kaplan-Meier analysis showed that the 5-year overall survival rate and 5-year disease-free survival rate of patients with high expression group and low expression group were 42.0% vs 66.9%(P=0.001)and 40.6% vs 65.4%(P=0.001).4. Multifactor analysis of Cox proportional hazard model found that the expression level of Zfx,lymph node metastasis and TNM stage(P=0.047,P=0.039,P=0.044) were independent prognostic factors.Conclusions1. The expression level of Zfx was significantly higher in colorectal cancer issues than adjacent normal tissues,and associated with tumor differentiation, lymph node metastasis and TNM stage,all of this prompt Zfx high expression are inseparable with colorectal cancer.2. Kaplan-Meier analysis showed that the 5-year overall survival rate and 5-year disease-free survival rate of patients with high Zfx expression group were significantly lower than patients with low Zfx expression group. Multifactor analysis of Cox proportional hazard model prompt that Zfx expression level have a predictive value for the prognosis of colorectal cancer patients,and its may be an independent prognostic factors. |
