Objective: To discuss Effects of CXCR4 monoclonal antibody on the blood-retinal barrier and expression of vascular endothelial growth factor in diabetic rat retina.Methods : Selecting Sixty-four adult male SD rats and dividing into normal control group(CON group) and diabetic model group by randomly. injecting streptozotocin(STZ) to establish diabetic retinal(DM) animal model, the control group was injected sodium citrate buffer. After three months, the diabetic rats were divided into treatment group and untreated group. The vitreous cavity of treatment group was given the CXCR4 monoclonal antibody. The control group and untreated group were given saline. After a week, observing each retina changes by HE staining of retina and the blood-retinal barrier changes by Evans blue(EB) retinal spread sheet and EB content determination of retina, detecting the content of VEGF by ELISA.Results: The HE staining of retina shows that the retina of CON group have no change; In the un-treatment DM group, each retina structure become disorder, retinal endothelial tissues appear edema, the retina have a large number of new blood vessels and some Vascular bud nuclei break out of the basement membrane; Comparing the un-treatment DM group, the retinal conditions of treatment DM group have good turns, the retinal endothelial tissue edema is lightened and the amount of neovascular are reduced. In the un-treatment DM group, the VEGF level was(194.91±2.18)pg·ml-1, markedly higher than the CON group(122.43±1.62)pg·ml-1(P < 0.05) and the treatment DM group(143.65±1.46)pg·ml-1(P<0.05). The retinal stretched preparation with EB showed that the CON group was normal, and the un-treatment DM group had a lot of leakage of fluoresced, and more than the treatment DM group. The retinal EB content of CON group, un-treatment DM group, and treatment DM group were(12.56 ± 0.28) ng·mg-1、(33.91 ±2.69) ng·mg-1、(21.49±0.51)ng·mg-1, and the data had statistical significant(P<0.05).Conclusions: CXCR4 monoclonal antibody can inhibit the expression of VEGF and the formation of new blood vessels in diabetic rat retina, reducing the damage of blood retinal barrier. It will become into a new drug on treatment of Diabetic retinopathy.
