The Influence On Colorectal Cancer Sensitivity To Chemotherapy Drugs By Cancer Associated Fibroblasts

Objective:To study the influence on human colorectal cancer lovo cell line sensitivity to chemotherapy drugs by Cancer associated fibroblaststs, optimization the scheme of screening cancer chemotherapy drug.Methods:(1) Using tumor cells and normal fibroblasts were com-cultured to build cancer-associated fibroblasts, and then using immunofluorescence technology to validate.(2) MTT method is used to detect the impact of cancer associated fibroblasts on human colorectal cancer cell line lovo cell and the chemotherapy drug sensitivity.(3) MTT method was used to detect the effect of fresh colorectal cancer tissue extract to lovo cell lines and chemotherapeutic drugs sensitivity.Results :(1) immunofluorescence staining results showed that after cocultivation, fibroblasts express tumor related indicators α-SMA.(2) MTT method results show that the single lovo cell lines are sensitive chemotherapy drug capecitabine(79.09), fluorouracil(68.74%), Taxotere(53.12%), methotrexate(70.15).the cocultivation groups sensitive chemotherapeutic drugs oxaliplatin(63.19%), Irinotecan(58.75%), Capecitabine(52.55%),fluorouracil(56.79%), the difference is obvious.(3) MTT method shows that fresh colorectal cancer tissue extract and fibroblasts cocultivate to lovo cell sensitive chemotherapeutic drugs are oxaliplatin(59.00%),Irinotecan(64.36%),fluorouracil(54.83%), which has a great influence on sensitivity of chemotherapy drugs, but there are no tatistical significance between the co-culture group and fibroblasts.Conclusions 1.Colorectal cancer cell line lovo cell lines can make normal fibroblast cells into a tumor associated fibroblasts. 2.The tumor associated fibroblasts affect the lovo cell lines sensitivity to chemotherapy drugs. 3.Human colorectal cancer tissue extracts and tumor associated fibroblasts affected on the chemotherapeutic sensitivity of lovo cell line 4.Compared with the group of single lovo cell line,the results of tumor chemosensitivity test of the group of cancer-associated fibroblasts(CAFs) and lovo cell line are more close to the results of the experiments of the group of human colorectal cancer tissue extracts and cancer-associated fibroblasts(CAFs) and lovo cells.
