| Objective:To investigate the change and significance about the expressions of MUC2, MUC3 in experimental acute terminal ileitis.Methods:The Selected 40 healthy adult male mongrel dogs were randomly divided into, A (normal control group), B (high temperature and high humidity high blood glucose group), C (normal temperature and humidity normal glucose craniocerebral firearm injury group), D (normal temperature and humidity hyperglycemia craniocerebral firearm injury group), E (high temperature and high humidity normal glucose craniocerebral firearm injury group), F (high temperature and high humidity hyperglycemia craniocerebral firearm injury group), G (normal temperature and humidity high blood glucose group), H (high temperature and high humidity in normal blood glucose group) 8 groups,5 rats in each group, respectively 30min before the experiment begin,respectively measured and recorded rectal temperature, heart rate, respiratory frequency and other vital signs of each group of experimental dogs.When the experiment begin,give each dog Be Lu Mianning(0.08ml/kg)-be used to intramuscular anesthesia, Aã€Bã€Gã€H group treated without ang special conditions,but Cã€Dã€Eã€F group with craniocerebral firearm wound respectively. Then the experimental dogs fixed on the aseptic operation table (supine),executed tracheal intubation, cardiopulmonary resuscitation and other rescue measures immediately, and closely monitoring the dogs heart rate, rectal temperature and other vital signs,in addition to cardiac massage and artificial bladder assisted breathing, not do other processing.After maintaining 2 hours,do operation under aseptic conditions, obtaining experimental canine’s distal ileum(taken from the ileocecal valve about 5cm).Before embedded by paraffin, the tissue should be observed by naked eyes.Using the method of hematoxylin eosin staining (HE staining) to make some of the tissue into pathological section, and to observe the changes in the electronic and optical microscope;using conventional SP immunohistochemical method to detect the expression level about MUC2 and MUC3 of the terminal ileum tissues after modeling.Results:1.The change of vital signs of the experiment The vital signs of experimental canine remain stable at less for 30 minutes before the experiment started,Compared with group A(normal control group),the vital signs like temperature and heart rate,of B,C,D,E,F,G and H group,there is not any statistical significance (P>0.05). temperature, heart rate differences had no After 1 hour,the temperature of B, C, G, H groups,had difference has no statistical significance,compared with A (P> 0.05).but the temperature of D, E, F group compared with group A, was statistically significant different (P< 0.05);C, D, G, H group compared with group A,there was no statistically significant difference in heart rate (P>0.05).The heart rate of group A, B, E, F was statistically significant (P<0.05).After 2 hours,the temperature of B, G, H group compared with group A,has no statistical significance in difference(P>0.05).However,the difference temperature of C, D, E, F group compared with group A,there was statistically significant (P<0.05);there was no statistically significant difference between group A,G,C,D(P>0.05),the heart rate of B, E, F, H group,compared with group A,there was statistically significant in difference(P<0.05).2.The change of terminal ileum tissue under macroscopic and the naked eye view As we can see of our eyes,the A, G, H group was not any obviously inflammatory change.But the B, C, D, E, F group all in acute inflammatory reaction for performance, and the inflammatory reaction of group F is the most severe than the other group. Under light microscope, A, G, H were no significant inflammatory changes, B, C, D, E, F groups have varying degrees of inflammatory cell infiltration, lymphatic, lacteal expansion, villi became shorter and thicker, the manifestations of acute inflammatory reaction.Two kinds of observation method of inflammatory score comparison between groups that craniocerebral firearm injury and high temperature and high humidity on the degree of inflammation of the distal ileum have effect, and the effect of hyperglycemia on the degree of inflammation of the distal ileum is not clear.3.The expression level of MUC2 in terminal ileum tissue The positive expression of MUC2 showed Brown granular staining is widely distributed in the cytoplasm of goblet cells,with perinuclear and nuclear membrane, obviously, a diffuse positive in cytoplasm, not in nucleus.The average optical density value of the expression of terminal ileum tissue MUC2 of each experimental group,compared with the A group,there were statistically significant differences between the groups (P<0.05); The comparison between groups about the average optical density value of the expression of MUC2 indicated that craniocerebral firearm in high temperature and high humidity have some effects on the expression of MUC2,but for hyperglycemia is not clear.There was a negative correlation between the degree of inflammation and the expression of MUC2 at the end of ileum.4.The expression level of MUC3 at terminal ileum tissue The positive expression of MUC3 showed Brown granular coloring is widely distributed in the cytoplasm of goblet cells and columnar cells, not in nucleus.Compared B, C, D, E, F, H group with A group,there were statistically significant (P<0.05), however,G group and A group had no significant difference (P>0.05);4.The comparison between groups about the average optical density value of the expression of MUC3 indicated that craniocerebral firearm in high temperature and high humidity have some effects on the expression of MUC3,but for hyperglycemia is not clear.There was a positive correlation between the degree of inflammation and the expression of MUC3 at the end of ileum.Conclusions:1.Craniocerebral firearm injury can cause inflammation of the terminal ileum tissue changes, inflammatory injury aggravated with the superposition of high temperature and high humidity, high glucose condition;2The down-regulation of MUC2 and up-regulation of MUC3 at terminal ileum mucosa were related with the degree of ATI inflammatory injury,.Mucin secretion may be involved in the pathogenesis of experimental ATI. |
