The main purpose of this paper is to sift the hypoglycemic activity part from the powder of Hericium erinaceus (the powder extracted from Hericium erinaceus deep submerged fermentation culture), to make further confirmation on the hypoglycemic pharmacodynamics and to investigate the mechanism of hypoglycemic activity.80a is a homogeneous relative molecular mass polysaccharide, which was isolated from the bioactive part. This study could elucidate preliminarily the fundamental substance of the hypoglycemic activity in the powder of Hericium erinaceus and will be helpful for the further development. The main research contents and results are as follows:1. The anti-hyperglycemic screening of the isolated major fractions from the powder of Hericium erinaceus.The probable compounds containing in the powder of Hericium erinaceus were systematically investigated by chemical preliminary tests. The results indicated that the powder of Hericium erinaceus including proteins, organic acids, saccharides, volatile oils, phenols and lactones.The95%ethanol extracts, the crude polysaccharide and water soluble extracts were used to screen the isolated major compounds from the powder of Hericium erinaceus. Select the hypoglycemic effective parts by each group’s fasting blood glucose after21days of intragastric administration. Comparing to control group, the FBG of the Type2diabetic mouse which administrated with the polysaccharides from the powder of Hericium erinaceus dropped significantly(P<0.01). It is concluded that the crude polysaccharide is the anti-hyperglycemic active fraction.2. The extraction technology and the assaying of the HEP.Single-factor test combined with orthogonal test were applied to optimize the extraction technology of the polysaccharides from the powder of Hericium erinaceus. The optimum extraction technology of polysaccharides from the powder of Hericium erinaceus:impregnating with stilled water for15minutes; refluxing and extracting twice at70℃;2hours each time, concentrated to4times dosage of crude drug (g/mL); alcohol precipitation concentration was80%. Under the optimum technological conditions, the polysaccharides yield was up to22.72%.Phenol-sulphuric acid method is used to determine the sugar content of the crude polysaccharides (24.29%) and the refined polysaccharides (48.08%).The Bradford method is used to determine the protein content of the crude polysaccharides (4.94%) and the refined polysaccharides (2.63%).3. The hypoglycemic activity of the polysaccharides from the powder of Hericium erinaceus.Different diabetic models are adopted to validate the hypoglycemic activity of the polysaccharides from the powder of Hericium erinaceus. The polysaccharide shows the ability of resisting the diabetes mellitus (DM) induced weight-loss, lowering FBG,adjusting the confused blood fats, protecting the viscera and improving glucose tolerance in DM mice.4. The separation, purification, physico-chemical property and structural analysis of the polysaccharides from the powder of Hericium erinaceus.To isolate and purify the polysaccharides from the powder of Hericium erinaceus then study their physical-chemical property, conformation and construction. The polysaccharides were graded to HEJP30, HEJP60and HEJP80by ethanol. DEAE column chromatography and SephadexG-100column chromatography are applied to further purification.80a is a homogeneous relative molecular weight polysaccharide, which was obtained in the experiment. The purity, conformation and molecular weight of polysaccharides were evaluated by HPLC-ELSD gel exclusion chromatography.80a is a glucosans whose molecular weight is3487Da.
