Objective This study aims to explore the level of IL-17and IL-10and their function in serumof patients with hepatic CE in different cyst stages and to investigate the usefulness of serumcytokine dosage in the clinical diagnose of cystic echinococcosis (CE) in different cyst stages.To explore the immunological regulation mechanism of mice infected with Echinococcusgrannulosus,and to provide the evidence to enhance the theory of immunological escape inEchinococcus granulosus. Methods33patients with hepatic hydatid disease and21healthypeople(control group)were enrolled in this study. Sera divided into4groups. Patients withCE1-2(active, n=14), CE3(transitional, n=10),and CE4-5(inactive, n=9) and healthyindividuals (control group, n=21), The serum concentrations of IL-17and IL-10weremeasured by enzyme-linked immunosorbent assay.All data were analyzed by one wayanalysis of variance.LSD—t test analysis and Pearson correlation analysis. The course of themice from Kunming germ-line,30mice were randomly divided into2groups,normal controlgroup and Echinococcus grannulosus infected group.2ml Echinococcus granulosus wereintraperitoneal injected into Echinococcus granulousus infected group to establishEchinococcus granulousus infected model.After20weeks,and then kill the2groups ofmice.Spleen cells from mice were collected and CD4+T cell and CD4+CD25+Treg cells weredetermined by flow cytometer Results Serum levels of IL-17were significantly higher inthe CE3(transitional) group[(56.61±6.19) pg/ml]compared with those in the CE1-2(active)group[(38.57±3.83) pg/ml] and CE4-5(inactive) group[(40.22±5.15) pg/ml],(P<0.05); Therewas no significant difference in serum IL-17levels between the CE1-2(active)[(38.57±3.83)pg/ml] and CE4-5(inactive)group [(40.22±5.15) pg/ml]. Serum levels of IL-10weresignificantly higher in the CE3(transitional) group[(14.65±2.15) pg/ml]compared with thosein the CE1-2(active) group[(11.38±0.71) pg/ml](P<0.05), Serum levels of IL-10in theCE1-2(active) group[(11.38±0.71) pg/ml] were also significantly higher than the CE4-5(inactive) group,(P<0.05); The Thl7/Treg(IL-17/IL-10) ratio was significantly decreased inthe CE1-2(active) group(3.44±1.04) compared with that in the CE4-5(inactive) group (5.39±1.95); There was no significant difference of Thl7/Treg(IL-17IL-10)ratio between theCE1-2(active)(3.44±1.04) and CE3(transitional)group(4.6±2.4);There was also nosignificant difference of Thl7/Treg (IL-17/IL-10)ratio between theCE3(transitional)group(4.6±2.4) and CE4-5(inactive) group(5.39±1.95). The serumconcentrations of IL-17were has no correlation with IL-10in in different cyst stages.Echinococcus granulousus infected group were11mice infected,through routine pathologicalsection could saw the integrity structure of stratum germinativum and stratum corneum. Thepercent level of CD4+CD25+T cell were significantly higher in the Echinococcus granuloususinfected group compared with normal control group.The ratio of CD4+CD25+/CD4+T cellwere also significantly higher in the Echinococcus granulousus infected group compared withnormal control group. Conclusion The synergy effect of IL-17and IL-10promote the parasitedeath process in the later stages of the infection, IL-17and IL-10level could not as a markerof biological activity of echinococcal cysts independently, but Thl7/Treg(IL-17/IL-10)ratioreflect the activity of hydatid cyst to some extent, but as a maker to detect different cyst stagesis still showed many limits. The integrity of the Hydatid internal capsule corneous layer andthe biomass of the parasite may play an important role in deciding the degree and orientationof host immune response;the Bi-regulation of IL-10play an important role to in thedevelopment of disease. CD4+CD25+T cell play an important role to maintain host immunesystem for an inhibitory state to be beneficial to proliferation of Echinococcus granulosus inhost.
