| Part I The expression of Pim-1in androgen independent prostate cancer animal modelObjective:To investigate the different expression of Pim-1at gene and protein level in the transition from androgen dependent prostate cancer to androgen independent prosate cancer in animal model.Methods:Using human prostate cancer cell lines LNCaP cell, we set up androgen dependent and androgen independent orthotopic prostate tumor animal models with BALBc-nu mouse, first we use gene trip technology to find the different expression genes, then we use RT-PCR, QPCR and immunohistochemistry to test the expression of Pim-1at mRNA and protein level about the two different tissues.Results:(1) gene chip results:Affymetrix microarray detection of tumor tissue showed that expression of Pim-1at gene level in AIPC tissues is higher than that in ADPC tissues, the difference ratio is2.30771; the expression of Pim-1in ADPC tissues and castrated3days has not significant difference.(2) RT-PCR results:The result of agarose gel electrophoresis showed that the expression of Pim-1between AIPC tissues was higher than ADPC tissues(the difference has statistically significant, P<0.01), the difference ratio is1.93; the expression of Pim-1in ADPC tissues and castrated3days tumor tissues has not significant difference(P>0.05), it was consistent to the gene chip results.(3)QPCR results:Using2-△△Ct relative quantification method to analysis data, the expression of Pim-1mRNA level in AIPC tissues is higher than that in ADPC tissues(the difference has statistically significant, P<0.01), the difference ratio3.01; the expression of Pim-1mRNA level in castrated3days tumor tissues is also higher than that in ADPC tissues(the difference has statistically significant, P<0.05), the difference ratio2.05, it was nearly consistent to the gene trip and RT-PCR results.(4)Immunohistochemical results:Using Image-Pro Plus (IPP) image analysis software on immunohistochemical image analysis. The expression of Pim-1protein level in AIPC tissues is higher than that in ADPC tissues(the difference has statistically significant, P<0.01), the expression of Pim-1protein level in ADPC tissues and castrated3days tumor tissues has not significant difference(P>0.05), it was consistent to the above results.Conclusions:The expression of Pim-1in androgen independent prostate cancer is higher than that in androgen dependent prostate cancer, Pim-1may play an important role in the transition from androgen dependent prostate cancer to androgen independent prosate cancer.Part â…¡ The expression of Pim-1in human androgen independent prostate cancerObjective:To investigate the different expression of Pim-1at protein level in the transition from androgen dependent prostate cancer to androgen independent prosate cancer in human prostate cancer tissues.Methods:The specimens are selected from10cases of surgical resection specimens from the second hospital of tianjin medical university from January2009toDecember2010(8BPH patient,5AIPC patients and5ADPC patients,4prostate cancer patient after ADT therapy), diagnose by clinical datas and pathology. Using the different specimens to make paraffin sections, then we use immunohistochemistry to test the expression of Pim-1ã€AR about the two different cancer tissues and use Image-Pro Plus (IPP) image analysis software on immunohistochemical image analysis.Results:(1) The expression of AR at protein level in AIPC tissues is higher than that in ADPC tissues(the difference has statistically significant, P<0.01).(2) The expression of Pim-1at protein level in AIPC tissues is higher than that in ADPC tissues(the difference has statistically significant, P<0.01); the expression of Pim-1at protein level in BPH tissues is lower than that in ADPC tissues.(the difference has statistically significant, P<0.01); the expression of Pim-1at protein level between ADT therapy prostate tumor tissues and ADPC tissues has no statistically significant, P>0.05. It was consistent to the animal model results.Conclusions:The expression of Pim-1in human androgen independent prostate cancer is also higher than that in androgen dependent prostate cancer. Pim-1may play an important role in the transition from androgen dependent prostate cancer to androgen independent prosate cancer. |
