| Objective:To explore the magnesium isoglycyrrhizinate injection of jaundice obstructive liver injury protection.Methods:Choose only32healthy male SD rats, weight (200±20) g, which are supplied by the animal experiment center of the second military medical university; Raising the rats in an environment with a temperature of25to28℃and a humidity of60%~80%. We randomly divided the32rats into4groups of8rats each. It includes biliary obstruction model+magnesium isoglycyrrhizinate injection treatment of routine dose group (group A); biliary obstruction model+maximum dose of magnesium isoglycyrrhizinate injection treatment group (group B); biliary obstruction model blank control group (group C) and sham operation group (group D). Model:group A and group B and group C go ligation common bile duct respectively and reoperate after8days, inserting infusion catheter to the dilated common bile duct that due to obstruction, ligation done and purse string suture into the small intestine on the other side. Group D only make free bile duct ligation. Group A do the intraperitoneal injection of magnesium isoglycyrrhizinate of30mg/kg; group B go with intraperitoneal injection magnesium isoglycyrrhizinate of60mg/kg, group C and group D with0.9%Physiological saline injection by intraperitoneal injection of30mg/kg. Respectively in each1,3,5,7days after surgery we collect a1.5ml of blood in rat eye socket,isolate the serum and check the serum liver biochemical indexes with automatic biochemical analysis instrument Alanine aminotransferase (Alt), Aspartate aminotransferase (Ast), Total bilirubin (TBil), Direct bilirubin (DBil), Alkaline phosphatase (ALP), Γ-glutamine ammonia transpeptidase (GGT), ELISA-enzyme-linked immune sorbent assay to detect inflammatory factor:in the detection of peripheral blood interleukin1(IL1), interleukin6(IL-6), tumor necrosis factor alpha (TNF alpha) and nitric oxide synthase (iNOS). The results apply SPSS software for statistical analysis, measurement data were expressed as mean±standard deviation of each group and compared using ANOVA, groups were compared using LSD pairwise comparison.Results:The liver function change of rat serum:ligation of postoperative day1, group A and group B and group C serum ALT, AST levels were significantly higher than those in group D (P<0.05). Postoperative day3, group A and group B ALT and AST levels were significantly lower than that of group C (P<0.05). ALT and AST level between group A and group B with no statistically significant difference (P>0.1).After the2operations of the third day, groups of ALT and AST decreased to normal, no significant differences between groups (P>0.1). Group A and B than in group C in the rat serum TB, DB, no significant difference (P>0.05). Rats serum inflammatory factor function changes: inflammation due to subclass indicators no significant difference (P>0.1).Conclusion:Magnesium isoglycyrrhizinate injection can effectively reduce the jaundice obstructive serum transaminase level and has obvious protective effect on liver function. |
