| ObjectiveStudy on different expression correlation between Spleen-Kidney Yang Deficiency and Heat-toxin Stagnation syndrome of AIDS to explore the mRNA expression profiles and nature of AIDS with Spleen-Kidney Yang Deficiency and Heat-toxin Stagnation syndrome,provide a objective basis of differentiation in clinic.MethodsApplying gene chip technology to test AIDS patients with syndrome of Spleen-Kidney Yang Deficiency and Heat-toxin Stagnation, observe the differential expression between the syndromes. All date were analyzed with the spss19.0software. Meanwhile using S AS system to select the mRNA and functional analysis.Results1.Compare with control group,73differently expressed genes were found in Spleen-Kidney Yang Deficiency group;34genes were up regulation.39genes were down regulation.The differential expression of gene may related to some biological basis such as:Selective expression of chemokine receptors during T-cell polarization〠TNF/Stress Related Signaling. The regulatory gene of Selective expression of chemokine receptors during T-cell polarization(IL12RB1) is up regulation, The regulatory gene of TNF/Stress Related Signaling(MAPK14) is down regulation.2.Compare with control group,197differently expressed genes were found in Heat-toxin Stagnation syndrome group,65genes were up regulation,132genes were down regulation.The differential expression of gene may related to some biological basis such as:Arachidonic acid metabolism, IL-2Receptor Beta Chain in T cell Activation, Taurine and hypotaurine metabolism. The regulatory genes of Arachidonic acid metabolism and Taurine and hypotaurine metabolism are GGT1(down regulation and GGT6(up regulation); The regulatory genes of IL-2Receptor Beta Chain in T cell Activation are BCLXL and SOCS3(up regulation)Conelusions1. Spleen-Kidney Yang Deficiency syndrome of AIDS is closely related to the genes of IL12RB1(up regulation) and MAPK1(down regulation).The paths of Selective expression of chemokine receptors during T-cell polarization which is controlled by the gene named IL12RB and TNF/Stress Related Signaling which is controlled by the gene named MAPK14may be the biological basis of Spleen-Kidney Yang Deficiency syndrome of AIDS.2.Heat-toxin Stagnation syndrome of AIDS is closely related to the genes of GGT1(down regulation),GGT6(up regulation),BCLXL(up regulation) and SOCS3(up regulation).The paths of Arachidonic acid metabolism and Taurine and hypotaurine metabolism are controlled by the genes named GGTland GGT6, and IL-2Receptor Beta Chain in T cell Activation which is controlled by the genes named BCLXL and SOCS3may be the biological basis of Heat-toxin Stagnation syndrome of AIDS. |
