Experimental Study On The Pharmacodynamics Of Xiongzhi Zhitong Capsule In Treatment Of Migraine

Objective:In this study, we aim to investigate the effect and possible mechanism of Xiongzhi Zhitong Capsule against migraine.Methods:1. The anti-migraine effect of Xiongzhi Zhitong Capsule was studied with migraine rat models caused by injection of nitroglycerin (NTG) subcutaneously and migraine mice models caused by injection of reserpine subcutaneously.2. The effect of promoting blood circulation of Xiongzhi Zhitong Capsule was studied with auricle microcirculation disfunction mice models and blood stasis rat models.3. The analgesia effect of Xiongzhi Zhitong Capsule was studied with hot plate inducing algesia and acetic acid inducing writhing response.4. The effect of sedative of Xiongzhi Zhitong Capsule was studied with locomotor activity frequency of mice, sodium pentobarbital inducing extended sleeping time in mice and sodium pentobarbital inducing sleeping amount of mice.Results:1. The Xiongzhi Zhitong Capsule can significantly regulate the contents of active substances in plasma and brain neurotransmitter of migraine rat model caused by injecting NTG subcutaneously. The contents of5-HT, CGRP, NE, ET in plasma and that of NE, DA in the brain tissue was significantly decreased, and the content of5-HT in the brain tissue rose after treating with Xiongzhi Zhitong Capsule. To the reserpine induced migraine mice, Xiongzhi Zhitong Capsule can also significantly rise the contents of5-HT in serum and brain tissue, reduce the content of DA in brain tissue.2. Xiongzhi Zhitong Capsule can significantly expand the micro-artery diameter, accelerate the microvascular flow velocity and increase the amount of open capillaries of auricle microcirculation disfunction mice, significantly reduce whole blood variability1and5, plasma viscosity, erythrocyte aggregation index, viscosity of the low-cut and high-cut of blood stasis rat mode.3. Xiongzhi Zhitong Capsule can significantly elevate the pain threshold in mice in the hot plate test and can significantly reduce the writhing amount caused by injecting intraperitonealy the glacial acetic acid.4.The locomotor activity frequency of mice during five minutes was reduced, the sleeping time with threshold dose of sodium pentobarbital in mice and the sleeping mice amount with sudbissociative dose of sodium pentobarbital was increased after treating with Xiongzhi Zhitong Capsule.Conclusions:Xiongzhi Zhitong Capsule could promote blood circulation, analgesia, anti-migraine which achieved mainly by regulating the level of vasoactive substances and brain neurotransmitters, improving microcirculation and hemorheology.
