"WEI" is one of the important processed methods in Radix Dolomiaeae. The WEI and raw products have been recorded by Chinese pharmacopoeia. However, according to the Chinese pharmacopoeia, there are no any differences in functions and applications between raw products and processed products. Therefore, this thesis just focus on this problem and research the machinism of processed from two angles, the rat intestinal absorption dynamics and the biotransformation degradation dynamics in vitro. It is also a significant method to support the research of Radix Dolomiaeae in clinic practices and pharmaceutics.In the research of comparing the differences between the raw Radix Dolomiaeae and processed Radix Dolomiaeae in the rat intestinal absorption dynamics. Building the situ perfusion model in rats and utilizing the detection method by HPLC-UV so as to acquire the surplus of Costunolide and Drodehydrocostus Lactone both are the key active components.Analyzing the parameters of the test, the research result shows that all of the intestinal segment including duodenum, jejunum and ileum show the good ability in absorbing the key components in raw Radix Dolomiaeae and processed Radix Dolomiaeae. Analyzing by the software SPSS, the result shows that the absorption of Costunolide and Dehydrocos in the raw products and processed products has no statistical difference in different test intestinal segments. In the raw products, the result of the absorption of Costunolide is:duodenum> ileum> jejunum; the result of the absorption of Dehydrocos is:duodenum> ileum> jejunum.In the processed products, the result of the absorption of Costunolide and Dehydrocos both are:duodenum> ileum> jejunum. All of the evidences indicate that duodenum is the best segments in the absorption of Costunolide and Drodehydrocostus Lactone.In the research of comparing the differences between the raw Radix Dolomiaeae and processed Radix Dolomiaeae in the biotransformation degradation dynamics in vitro. Preparating the different cncubation solutions, including the different pH value buffered solution, artificial gastric juice and intestinal juice, liver homogenate, urine, blood plasma, and adding the solutions of raw products and processed products in the cncubation solutions. Then sampling as the different timings and utilizing the detection method by HPLC-UV so as to acquire the surplus of Costunolide and Drodehydrocostus Lactone.The result shows that two key components in raw Radix Dolomiaeae and processed Radix Dolomiaeae are stable in six different cncubation solutions, including the buffer solution of pH value of7.4, the buffer solution of pH value of9.0, artificial intestinal juice, liver homogenate, urine, blood plasma. In addition, Dehydrocostus Lactone both in the raw and processed Radix Dolomiaeae are also stable in the artificial intestinal juice and artificial intestinal juice without pepsase.However, Costunolide both in the raw and processed Radix Dolomiaeae are not stable in the artificial intestinal juice and artificial intestinal juice without pepsase,and shows the obvious degradation. The degradation situation accords with the first order kinetics equations. Derecting the half-time (t1/2) of Costunolide in the artificial intestinal juice and artificial intestinal juice without pepsase, the result shows that the t1/2of Costunolide in raw Radix Dolomiaeae in the artificial intestinal juice and artificial intestinal juice without pepsase are11.456±1.121min,10.036±1.204min respectively and the t1/2of Costunolide in processed Radix Dolomiaeae in the artificial intestinal juice and artificial intestinal juice without pepsase are8.346±0.982min,7.565±0.886min respectively. The data declare that acid is the important factor in effecting the stable of Costunolide both in the raw and processed Radix Dolomiaeae. Analyzing by the software SPSS, the result shows that the degradation situation of Costunolide in the raw products and processed products has statistical difference in acid solution which suggests that Costunolide is more stable in raw products than processec products in acid solution.The innovation in studying Radix Vladimiriae is to combine processing with absorption and metabolism in vitro, and the significance of this thesis have two aspects:firstly, in the study of aborption, all of the evidences indicate that duodenum is the best segments in the absorption of Costunolide and Drodehydrocostus Lactone. Combining with the results of advanced research, this thesis has found out the mechanism of WEI in Radix Vladimiriae. Secondly, in terms of metabolism in vitro, this thesis has indicated the metabolism of key components in different solutions which relating with the metabolize.
