| Objective:Through the observation of the influence of Zhizhu pill on spleen deficiency of slow transit constipation intestinal transmission function of mice and the interstitial cells of Cajal C-kit, and SCF expression to investigate the possible mechanism of Zhizhu pill regulating intestinal movement.Methods:60SPF level Kunming mice were randomly divided into the normal group (10rats) and model group (50rats), after the success of modeling, the model mice were randomly divided into model group,high dose Zhizhu pills and low dose group, Maren pill group, mosapride group,10rats in each group, model sixteenth day after administration of7d, mice in each group to detect a general behavioral characteristics, the first time they defecated the black grains,6h defecation quantity and quality, the xylose content and colon mucosa pathological characteristics of serum; Immunohisto chemistry technique and Real-time PCR method detected the expression of C-kit and SCF’s protein and mRNA.Results:(1) By comparing with the normal group and model group, the time of first bowel movement prolonged significantly,the quality and quantity of6h defecation reduced obviously,and serum xylose content decreased evidently,the epression of C-kit and SCF’s protein and mRNA decreased sharply (P<0.01).(2) By comparing with the model group and Mosapride group, high dose Zhizhu pills and low dose group show significant differences:the time of first grain of black out is obviously shortened; the quantity and quality of6h defecation increased evidently (P<0.01),the serum D-xylose content increased significantly (P<0.01);the comparison between two groups in statistics indicate significant differences (P<0.01or P<0.05). Compared with Maren capsule group, low dose group show that:the time of Zhizhu pill first bowel movement is shortened, the quality and quantity of6h defecation decreased (P<0.01),no significant difference of high dose Zhizhu pills group in statistics (P>0.05);serum xylose content of Zhizhu pills high dose and low dose group increased obviously (P<0.01).(3)By comparing with the model group, Maren capsule group, Zhizhu pills high dose and low dose group of C-kit protein and mRNA expression was increased remarkblely (P<0.01), no statistically significant difference between the two groups (P>0.05). By compared with Mosapride, Zhizhu pills high dose and low dose group of C-kit Protein and mRNA expression indicate no significant difference (P>0.05).(4) By comparing with the model group, Maren capsule group, high dose group of Zhizhu pill, SCF protein and mRNA expression was significantly increased (P<0.01).the comparsion between the two group show no significant difference in statistic (P>0.05). By comparing with Mosapride, Zhizhu pill in low dose group, SCF protein and mRNA expression increased obviously (P<0.01);and no significant difference between the high dose group of Zhizhu pill in statistic (P>0.05).Conclusion:Pathogenesis of slow transit constipation is relate to decreasing about the levels of the Colonic mucosa C-kit and SCF’expression.Zhizhu pills can treat spleen deficiency constipation through up regulating expression of C-kit, SCF in order to improve the quantity and distribution of interstitial cells of Cajal.The experiment indicate that low dose Zhizhu pills group is the best way to treat spleen deficiency of slow transit constipation. |
