Objective To explore the expression changes of islets cells and immune cells whichinfiltrated the islets, and cytokines changes followed by. We observed the changes of islet amyloidpolypeptide (IAPP), somatostatin (SS), CD68and CD22positive cells in islets of type1diabetesmellitus (T1DM) mice, and serum levels of interleukin-12(IL-12) and interleukin-4(IL-4).Methods104male C57BL/6J mice were divided into a normal control group(NCG), a salinecontrol group (SCG) and a experimental group(EG). We established the diabetes model in mice bylow-dose streptozotocin(MLD-STZ) injection (40mg/kg body weight for5days). The excisions ofthe pancreas tails removed on the3days,7days,10days,14days,21days and28days. Tissues wereassessed by immunohistochemical SABC, immunofluorescence, enzyme linked immunosorbentassay (ELISA), image analysis and morphmetric methods in the study. Results1. As comparedwith NCG and SCG, The fasting blood-glucose of T1DM mice rose since the14th, and rise to thetop at28th(P<0.05).2. Both of the number and area of islets in EG decreased.3. The numericaldensity on area of IAPP positive cells decreased since the3thday, but the immunoreaction ofIAPP-IR cells in EG increased, the mean density of IAPP-IR cells in EG increased since the7thday (P<0.05).4. As compared with the NCG and SCG, the numerical density on area of SSpositive cells increased since the3thday, the immunoreaction of SS positive cells in EGincreased, and the mean density of SS positive cells in EG increased since the7thday (P<0.05).5.The results of immunofluorescence double staining showed that, the IAPP and SS couldcoexpression in part of cells in islets.6. As compared with NCG and SCG, numerical density onarea of CD68positive cells maintained at a high level during the study.7.The numerical density onarea of CD22positive cells increased since the3th, but reduced after the7th.8. ELISA showed thatIL-12in serum of T1DM mice increased since the7thday, and maintained at a high level duringthe study, but IL-4level decreased, as compared with NCG and SCG (P<0.05). Conclusions1.The area and number of islets decreased in type1diabetes.2. The number of IAPP-IR cells decreased, but the immunoreaction of IAPP increased. The immunoreaction and number of SS-IRcells increased, and IAPP and SS coexpression in part of cells in islets, both of them involved thepathogenesis of type1diabetes.2. CD68and CD22positive cells infiltrated islets at the early stageto elevated the type1diabetes by antigen presentation.3. The serum IL-12increased and IL-4decreased in EG, suggested that Th1/Th2cells lose the balance.
