Background:Acute myocardial infarction (AMI),a worldwidecardiovascular disease with high morbidity and mortality,isthe acute necrosis of myocardial tissue due to persistent andsevere ischemia. In recent years, increasing evidence suggeststhat non-coding RNAs play important roles in pathophysiologyof AMI. Of them, micro RNAs (miR) as biomarkers of AMI have beenlargely investigated. For example, miRNAs such as miR-1,miR-499and miR-208were increased in serum of patients withAMI,suggesting miRNAs could be used as potential biomarkersand therapeutic targets of acute myocardial infarction.However,little is known about other classes of ncRNAs in this field.Long non-coding RNAs (lncRNAs), classified as transcriptslonger than200nucleotides, function in a wide range ofprocesses and can regulate gene expression by diversemechanisms.Lnc RNAs in fluids are relatively more stable as they often form secondary structures.The whole genometranscriptome analysis show that, after myocardial infarction,a large number of cardiac specific lnc RNAs have uniquefunctional properties and regulation for myocardial remodeling,myocardial regeneration, poor heart function, meaning cardiacspecific lnc RNAs have sufficient possibility to be as heartdevelopment and disease related molecular target andbiological markers. Consistently, the circulating longnon-coding RNA LIPCAR was explored as prognostic biomarkers forheart failure.However, study about the circulating longnon-coding RNA as biomarker of AMI is limited.A recent study found a lncRNA named UCA1(Urothelialcarcinoma associated1urothelial carcinoma,1), which ishighly expressed in the heart tissue, suggesting that it maybe a biological marker for cardiovascular diseases.The UCA1originally was identified in human bladder transitional cellcarcinoma and as the biomarker could be detected in the urineof patients with bladder carcinoma.In normal states, UCA1onlytranscribed at high levels in placenta, embryo, and most offetal tissues, but its expression was turned off in most tissuesafter birth.In adult, UCA1was only expressed in heart, spleenand its level was higher in heart, indicating it might be as novel biomarker of AMI.Objective: To evaluate the potential role of UCA1in thediagnosis and treatment of acute myocardial infarction, we testthe expression level of lncRNA-UCA1in plasma of patients withacute myocardial infarction and healthy people.Methods: Subjects: We selected49patients with acutemyocardial infarction (AMI) and15healthy people. Accordingto the time after acute myocardial infarction, the patientswith AMI in our hospital from2014November to2015Januarywere divided into7groups:0-2hours (2cases),2-6hours (3cases),6-12hours (6cases),12-24hours (12cases),24-48hours (13cases),48-72hours (9cases)and72-96hours (4cases).To investigate the effect of UCA1in the diagnosis andtreatment of acute myocardial infarction, we tested theexpression level of lncRNA UCA1in plasma of patients with acutemyocardial infarction and normal control group and compared theexpression levels of UCA1at different time.Results:1、Compared with the healthy group, the expression levelof UCA1in the plasma of AMI patients was decreased and therewas statistical significance (P<0.05). However, the expressionlevel of UCA1was sligtly down-regulated or even up-regulated in some samples.2、The expression levels of UCA1in the patients with acutemyocardial infarction are altered at different time point afterAMI. The expression levels of UCA1in plasma of AMI patientswere decreased at2-48h after AMI,there was statisticalsignificance compared with healthy control group (P <0.05),of them, the level of UCA1was the lowest at6-12hours afterAMI;there were no statistical significance of the UCA1in0-2hand48-72h(P>0.05);the expression levels of UCA1in plasmaof AMI patients were rised at72-96h after AMI.3、There was no statistical significance (P>0.05) in theexpression level of UCA1among AMI plus hypertension anddiabetes group, AMI plus hypertension group, AMI plus diabetesgroup and AMI alone group.4、The expression level of UCA1is negatively correlatedwith the miR-1in plasma of patients with acute myocardialinfarction.Conclusion:1、The UCA1could be used as a potential biologicaldiagnosis marker and therapeutic target of acute myocardialinfarction.2、The expression level of UCA1is negatively correlatd with the miR-1in plasma of patient with acute myocardialinfarction.
