Background:In the acute stage of ischemic stroke or transient ischemic attack,patients have a higher risk of recurrent or symptoms worsen. Muchhas happened in the onset of48-72hours. Recombinant tissuetypeplasminogen activator (rt-PA) has been proven to be effective methodfor the treatment of ischemic stroke, but because ofits strict administration time window, its application is restricted.Therefore, the majority of clinical guidelines recommend for the majorityof patients with ischemic stroke should be given antiplatelettherapy. Aspirin and clopidogrel isclinical drug most commonlyused antiplatelet aggregation, its effects are morecommon in clinicaltrials, the observation index for the incidence of cerebrovasculardisease new events. Because of the large number of clinical observationof uncontrollable factors, the purpose of this experiment was to use ratfocal ischemia reperfusion model, and was in-depth studyof safety, effectiveness of different antiplatelet drug therapy. Objective:This study used the Zea-Longa methods for rat model of middlecerebral artery ischemia. Give the rats aspirin and clopidogrel byintragastric administration.Compare the dynamic change of thecognitive function,and nerve function score and Peripheral bloodtumor factor and interleukin dynamic change, to explore the effect oftwo kinds of drugs.Method:Preparation of rat middle cerebral artery ischemia model using250-300g SD rats with suture method. Sham operation group (groupA, n=6), control group (group B,n=6), aspirin group (group C, n=6), clopidogrel group (group D, n=6).From the day following the administration into the group, giveaspirin10.28mg/kg/d for gavaging, lasting21days. Give D groupeclopidogrel7.71mg/kg/d for gavaging lasting21days. Start line of watermaze experiment when gavaging for fourteenth days and,start the watermaze test in twenty-first day when Drug treatment lasts for seventhdays,and twenty-first days,we start nerve function score. use ELISA todetermine the change of tumor necrosis factor(TNF), interleukin1(IL-1),interleukin6(IL-6) in serum。.Results:1, neural function scores of clopidogrel medication rats are higherthan aspirin therapy group. 2, clopidogrel treated patients than aspirin medication group escapelatent period is short, and many times across the platform.3, the model group of peripheral blood TNF, IL were significantlyhigher than those in the sham operation. Clopidogrel medication groupand aspirin medication group compared with the control group, the serumlevels of TNF, IL significantly reduced.More pronounced resultspresented by clopidogrel medication group.Conclusion:1anti platelet aggregation drugs can improve the rat middle cerebralartery ischemia reperfusion model of cognitive function.2anti platelet aggregation drugs can inhibit the ischemiareperfusion inflammatoryreaction.3.Clopidogrel and aspirin are effective for ischemic stroke.
