Objectives:Through detect serum Nestin level in peripheral venous of severepreeclampsia (SPE),normal pregnancy(NP) and healthy nonpregnancy(HNP)women,to investigate the correlation between serumNestin level and age, gestational age, systolic pressure, diastolicpressure and mean arterial pressure (MAP), to provide monitoringindex for new vascular endothelial cells and experimental basis onpathogenesis of SPE.Materials and Methods:Collected serum from inpatients,prenatal examination and healthy nonpregnancy women in Jilin University Bethune Second Hospital from2012October to2014December.Using a standard sphygmomanometer to measure theblood pressure of right arm, with the mercury column was reducing, referedto the first sound pressure value was the systolic pressure, when the soundwas disappearing,the pressure value was diastolic pressure. Mean arterialpressure (MAP)=diastolic pressure+1/3pulse pressure.The object ofstudy: from84cases of SPE patients, including40cases of EOSPE and44cases of LOSPE;129cases of NP women,including45cases of normalpregnancy women in less than20W(NP-1),84cases of normal pregnancywomen were matched SPE in ages, gestational ages,normal blood pressuresand negative of urine protein.Among the84cases of normal pregnancywomen,41cases of EOSPE control(EOSPE-C) women matched the ages andgestational ages with EOSPE,there were43cases of LOSPE control (LOSPE-C)women matched LOSPE in ages and gestational ages;72cases of HNP women were normal pressure and negative of urine protein.Using enzyme-linkedimmunosorbent assay (ELISA) to detecte serum Nestin levels. SPSS21.0software was used for the statistical analysis of dates.Results:1.Serum Nestin levels in SPE was highest,the second group was SPEcontrol(SPE-C) women.The lowest group was HNP women. The levels of SPEin serum Nestin were higher than that of SPE conrol group, approachingstatistical significance(P=0.054).The serum Nestin levels of SPE groupwere significantly higher than HNP group(P=0.004),The serum Nestinlevels of SPE-C were higer than HNP, but there was no statisticalsignificance (P>0.05). In LOSPE group, the serum Nestin levels werehigher than that of EOSPE-C,approaching statisticalsignificance(P=0.051).The levels of serum Nestin in LOSPE group weresignificantly higher than that in HNP Group(P=0.002). In129casesof normal pregnancy women with gestational age5.4W~40.7W,the serumNestin levels were higer than that of HNP group(P=0.014).In NP group,45cases of gestational age <20W NP (NP-1)women’s serum Nestin levelswere significantly higher than gestational age≥20W in SPE control womenof84cases(P=0.001).NP-1women’s serum Nestin levels were verysignificantly higher than healthy non pregnancy women(P=0.000).2. In EOSPE, there was a positive correlation between the serum levelsof Nestin and gestational age (r=0.334,P=0.035).In NP group with age>25y≤35y, there is a very significantly negative correlationbetween the level of Nestin in serum and age (r=-0.454, P=0.000).Theserum Nestin levels of age>35y≤45y women in HNP group had aapproaching positive correlation with age(r=0.529,P=0.052).3. In EOSPE group,there was a very significantly negative correlationbetween the level of Nestin in serum and gestational age(r=-0.544, P=0.000),but LOSPE women’s serum Nestin levels and gestational agehad a significantly positive correlation (r=0.500, P=0.001).NPwomen’s had a significantly negative correlation between the level ofNestin in serum and gestational age(r=-0.283,P=0.001).In45casesgestational age at5.4W~19.9W NP-1women,the serum Nestin levels andgestational age showed a significantly negative correlation(r=-0.305,P=0.042).IN SPE group, gestational age≥20W<28W SPE-1womenhad the highest serum Nestin levels,the second one was gestational age≥37W<41W SPE-3group,the lowest group was gestational age≥28W<37W SPE-2.There was a significant statistic in SPE-1and SPE-2(P=0.000).SPE-2women’s serum levels of Nestin were significantlylower than SPE-3(P=0.001).The serum Nestin levels in SPE-1weresignificantly higher than SPE-1control(SPE-1-C)(P=0.001),SPE-3women’s serum Nestin levels were significantly higher than SPE-3control(SPE-3-C)(P=0.006). There was a positive correlation betweenserum Nestin levels and gestational age in gestational age≥28W<37W SPE-2women(r=0.308,P=0.029).4. The serum Nestin levels had a positive correlation with SP,DP andMAP in SPE group (r=0.316,P=0.037;r=0.386,P=0.010;r=0.402,P=0.007).It had obviously positive correlation with SP,DP and MAP inLOSPE(r=0.316,P=0.037;r=0.386,P=0.010;r=0.402,P=0.007).Theserum Nestin levels only had a negative correlation with SP in NP(r=-0.199, P=0.023).In EOSPE-C group,there was a approaching correlationbetween the serum Nestin levels and DP (r=0.304, P=0.053),andit has a positive correlation between the serum Nestin levels and MAP(r=0.321, P=0.041). The serum Nestin levels had a significantly negativecorrelation with SP,DP and MAP in LOSPE-C group(r=-0.414,P=0.006;r=-0.410, P=0.006;r=-0.422,P=0.005).In SPE group,≥28W<37W SPE-2patient’s serum Nestin levels had a positive correlation with SP,DP and MAP(r=0.317,P=0.025;r=0.409,P=0.003;r=0.389,P=0.005).There was a positive correlation between serum Nestin levelsand DP in≥20W<28W SPE-1control group of NP(r=0.485, P=0.019),its serum Nestin levels had a positive correlation with MAP(r=430, P=0.041).In≥28W<37W SPE-2control women(SPE-2-C),theserum Nestin levels had a approaching negative correlation with DP(r=-0.331, P=0.056).Conclusions:1.The increase of serum Nestin level is one of the characteristicsin SPE,especially in most LOSPE patients,it maybe a protectivemechanism.It maybe lack in most EOSPE patients.2.Age is a positive factor to the serum Nestin level of age groupsat35y~45y HNP women.It has a negative correlation with serum Nestinlevel in NP women with age groups at25y~35y and it is a positive factorin the serum level of SPE.3.Gestational age is the serum Nestin level’s negative factor inNP,especially in gestational age less than20W and it’s a positive factorin SPE≥28W<37W.Before28W and after37W are the important periodfor increasing serum Nestin level in SPE women.4.SP is a negative factor to serum Nestin level in NP,especially ingestational age>20W.Blood pressure is a positive factor to serumNestin level in SPE, especially in LOSPE patients.5.The serum Nestin level may be a index of proliferation in SPE newvascular endothelial cells.
