Study On The Mechanism Of The Effect Of Shenhong Buxue Granule On Hematopoietic And Immune Function Of Blood Deficiency Animal Model

Purpose：Observing the effect of Shenhong Buxue Granule on blood deficiency modelanimal’s hematopoietic function, immune function and correlation factor, which copied byacetylphenylhydrazine and cyclophosphamide, to investigate the mechanism of ShenhongBuxue Granule on improving blood deficiency, enrich the connotation and make furtherresearch on mechanism of Chinese traditional medicine’s prophylaxis and treatment theoryabout blood deficiency, provide new ideas and basis for drug development.Method: Subcutaneous Injection of acetylphenylhydrazine20mg kg-1at the second dayof the experiments, and40mg kg-1at the fourth day with the same method, and from thefourth day onwards, intraperitoneal injection of cyclophosphamide40mg kg-1,1times a dayand for4consecutive days, to replication the model of animal blood deficiency syndrome.Intragastric administration the rat of Shenhong Buxue Granule(9g· kg-1,6g· kg-1,3g·kg-1)for14days. Outcome Measures:(1) Using the blood cell to analysis and detects the changeof peripheral blood routine, and ELISA to detects the cell factors’change which are associatedwith hemopoiesis function in the animal’s serum(IL-3, EPO, G-CSF, M-CSF, VCAM-1).(2)Using ELISA to detects the cell factors’change which are associated with immune function inthe animal’s serum(CD3, CD4, CD8, IL-2,IL-6, TNF).Result:(1)According to the observation on the hematopoietic function in mice withblood deficiency and influence factors, Shenhong Buxue Granule can significantly increasethe number of peripheral blood RBC, PLT, and the content of HGB, HCT, It alsocan increase the cell factors (IL-3,EPO, G-CSF, M-CSF, VCAM-1) that associated withhematopoietic cytokines’expression levels in various degree;(2) The detections about the cellfactors that associated with immune function showed that Shenhong Buxue Granule canincrease the expression level of CD3, CD4,CD8, IL-2, IL-6, TNF, which are in the serum ofblood deficiency model mice.Conclusion: The experimental study indicated that,(1) Shenhong Buxue Granule cansignificantly promote the hematopoietic function of blood deficient model mice, and itsmechanism may have something to do with the drug regulator of hematopoiesiscytokine;(2) Shenhong Buxue Granule can obviously promote the immune function of blood deficient model mice, and its mechanism may be related to the drug regulation ofimmunization cytokines. However,The detailed mechanism needs further study.
