Objective: To study the effect of ropivacaine-induced toxicity on learning and memory abilitiesand the expression of GFAP in the hippocampus of immature rats.Methods: Sixty28-d-old SD mice were randomly divided into ropivacaine inducement group(Convulsion was defined as that it achieved Racine classification V) and sodium chloridetreatment group (n=30). Immature mice in each group were subdivided into4subgroupsaccording to the different injection times (24h,3,7and60d after convulsion orinjection,n=5);Meanwhile the expression of GFAP in the hippocampus at each time point werealso measured for immunohistochemistry.The left10mice in each group were performed Mirrorwater maze test to explore the latency in finding the platform according to at time(1~3d、5~7d、58~60d).Results: The mices’ mean escape latency of the ropivacaine inducement group were longer thanthat of the control group a24h and3day(P<0.05),but there was no significant differencebetween R and N at7day and60day.The number of GFAP-positive astrocytes in the ropivacaineinducement group were less than the sodium chloride treatment group(P<0.05), especially on the3day after ropivacaine-induced toxicity, with smaller cell body, thinner and shorterprcesses,stained more lighter.Conclusion: Ropivacaine-induced toxicity could resuld in cognitive impairment in the immaturerats for a short time,and could restrain the expression of the GFAP in astrocytes.
