| ObjectiveTo investigate the protection of SSTF in the Parkinson’s disease(PD) C57BL/6Jmiceinduced by1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP) about its’ behavior,the number of dopamine(DA) neurons in the substantia nigra and the level of MDA in theblood serum, and to explore its protective mechanism in the PD mice model.MethodsChoose eight-week-old to ten-week-old male C57BL/6J mice.This experiment needs32mices,and these mices were divided into4groups randomly:normal controlgroup,MPTP+normal saline(NS)group, SSTF+MPTP group,MPTP+SSTF group.Afterinjecting MPTP into the mice for3-5min every time,we should observe its’ behavior, makethe fremitus grade,do the traction test and note the tration time; After the last of druginjection,execute these mices on the second day,but before executing them,phlebotomizefrom its’ tail,exam the level of MDA by managing the enzyme-linked immunosorbent assay(ELISA)in the blood serum of PD mices; and collect brain sample of mices that includethe substantia nigra,observe the number and modality changes of neurons in the substantianigra by the toluidine blue staining.Results3-5min later after injecting MPTP into mices,then the symptoms were:slow-moving,tremor, piloerection, raising,hunching, step length was shortened,step width increase,instability of gait,the MPTP group symptom was the most severe,the next wereMPTP+SSTF group and SSTF+MPTP group. Compared with the control group,othergroups had statistical significance(P<0.05).Compared with the MPTP group,other groupshad statistical significance(P<0.05). But MPTP+SSTF group and SSTF+MPTP group hadno statistical significance(P>0.05). The traction test showed,every two groups hadstatistical significance(P<0.05)except that MPTP+SSTF group and SSTF+MPTP group.Toluidine blue staining experiment manifests that compared with the control group,other groups’ DA neurons decreased,density losed,the modality of the DA neurons were plumpand nissl body distribute asymmetryly.Also,DA neurons of MPTP group and their coresoberviously shrink.Measuring MDA concentration in the mice blood serum by the ELISAexperiment,compared with the control group,the level of MDA in the MPTP group micemodel blood serum clearly increased.Other statistic index simlar to above.ConclusionIt can lead C57BL/6J mice to appear Parkinsonism symptom by peritonealinjection.The number of DA neurons decreases and modality changes in the the substantianigra pars compacta of midbrain.Compared with the control group,the level of MDA in theblood serum increases because of injecting MPTP.Pretreat and post-treat of SSTF canprotect PD mice by the MPTP induced.But pretreat and post-treat of SSTF had nostatistical significance.About protection mechnism of SSTF on the PD mice, therelationship between the SSTF’s dose needs,dose time,dose way and no significantdifference warrants futher investigation. |
