Objective: To explore the role of Osteopontin (OPN) in invasive growth and metastasis in animalmodel of hepatic alveolar echinicoccosis, and observe the effect of exogenous anti-OPN antibodywith growth and development of E.multolocularis (Em).Methods: To establish the animal model of Em, mice were infected with20%Em suspension vialiver injection.120Meriones unguiculatus were divided into3groups:Model group: Eminoculation only, Experiment group: Em inoculation and treatment with anti-OPN antibody viatail vein, and Control group: Em inoculation and treatment with blood serum via tail vein. Themice from each group were sacrificed on day140,180,220,and,260respectively. Hepaticechinococcus cyst and metastasis tissue were observed in HE stain, and the expression levels ofMMP-2、TGF-β1were measured by immuno-staining.Result:,1There were significant difference between Experiment group and Control group, andExperiment group and Model group on day140,180and220group(P﹤0.05)in the cyst wetweight, there was no significant difference between Control group and Model group (P﹥0.05).2The thoracic lymph nodes metastasis rates were significantly different between experimentgroup and comtrol group、experiment group and model group on day140and100respectively(P﹤0.05),while there were no significant difference between Experiment group and Control groupand Experiment group and model(P﹥0.05).3Light microscopy in HE staining showed the pathological change of cyst layer was nosignificantly difference between experiment group and comtrol group、experiment group andmodel group on day140,180, and220respectively(P﹥0.05).4The immuno-staining showed that MMP-2andTGF-β1mainly distributed in the germinallayer, the inflammatory cells, and some hepatocytes with different levels. There was no significantdifference in the levels of MMP-2between all time points in all groups. The positive expressionlevels of TGF-β1were significantly different between Experiment group and other groups onday140and180(P﹤0.05),but on day220(P﹥0.05).Conlusion: MMP-2and TGF-β1responses as important mechanism of host defence against thehepatic alveolar hydatid.The anti-OPN antibodies can reduce the expression of TGF-β1,this maybe one of the mechanisms of the inhibition of the growth and metastasis of E.multolocularis..
