Laminine was firstly found in marine seaweed, it is a kind of amino acidderivative which has antihypertensive activity and can prevent hypertension andcerebral hemorrhage. Laminine is a kind of national first class new medicine which hasnot been listed at home and abroad at present. The content of laminine in the seaweedis rare, not only the extraction process is difficult but also the separation cost is high.Comply with the requirements of green chemistry, this paper used chemical synthesismethod to prepare laminine with safe and low toxic dimethyl carbonate as raw material,aslo used the response surface optimization method to explore the various factorsaffecting the synthesis reaction aimed to improve the lysine microelement chelate andlaminine preparation technology, to improve product yield and purity, and to eliminatethe safety hidden trouble, reduce pollution. Last but not least, the paper wishes offerthe majority of patients with high blood pressure the safer and more effectiveantihypertensive drugs.Using lysine hydrochloride and metal salts as raw material, this pape adopt themethod of metal chelate to protect amino selectively. Through the single factorexperiment results of the lysine zinc and lysine copper chelate preparation process,three factors which influent greatly on yield are the system of pH, reaction temperatureand time. And the response surface method is used to optimize the preparation processof zinc lysine, the best condition of amino masking reaction is: pH=7.5, thetemperature of the reaction is58℃, time is2hours, the maximum yield of92.12%.Second, this paper has explored respectively for high pressure and normalpressure methylation reaction of dimethyl carbonate. By contrast, the yield of highpressure methylation reaction is slightly greater, but it need more harsh reactioncondition. And the response surface method is used to optimize the reaction of highpressure methylation. The best condition of high pressure reaction: the reactiontemperature is160℃, stirring speed is380r/min, dosage of potash is3.8%, thebiggest product yield was45.75%.Finally, the paper comprehensively used distillation, extraction, ion exchangecolumn chromatography and crystallization for isolation and purification of theproduct and explored the best separation condition of ion exchange columnchromatography. The yield of separation experiments under optimal conditions was82.25%.And the paper used the chromogenic reaction, TLC, HPLC, LC-MS, IR technique for identification of the laminine.
