| Object:The previous reseach hinted that pattern recognition receptors play an role in the pathogenesis of rheumatoid arthritis(RA) and sinomenine can inhibit the expression of downstream signal molecules and the transduction of signal which are belong to pattern recognition receptor,but the mechanism has no in-depth study. The subject posits that sinomenine cured RA by impact on the pattern recognition receptor. Through the study of organic tissue and related antibodies to research for the pattern recognition receptors which sinomenine may affect and the new mechanisms which sinomenine effect on the immune system, in order to provide laboratory evidence for clinical application of sinomenine.Methods:SD rats110, male and female, randomly we selected16for the control group, then we used the remaining rats to build arthritis rat model induced by mycobacterium tuberculosis(MIA) according to the literature.After model were completed,we randomly divided model group into the model control group,positive drug group, sinomenine high, medium and low treatment groups.We measured the weight and the modeling injected side foot(left)ankle circumference and paws thickness of rats periodically.we detected rat peripheral RF, IgG, IgM levels; we make pathorphological observation of rat’s synovial tissue of ankle joint(HE Staining)and we detected the expression of Mannose receptor which were come form rat’s spleen.Finally we analyzed all date with statistical software of spss.Result:1Sinomenine could effectively restrain the weight growth of rheumatoid arthritis rat model was slowed compared with control group;2Sinomenine could reduce the expression level of mannose receptor in rat spleen and related antibodies levels,thereby immune response were suppressed and synovial injuried, joint bone destruction were reduced;3the middle treatment sinomenine group improved significantly synovial and bone destruction, related indicators expression also changed;4rat’ankle circumference and paws thickness of each treatment group had no obvious improvement after treatment and the change trend was consistent with the model group.This trend could be considered as a certain self-healing in adjuvant-induced arthritis model rats.Conclusion:Sinomenine could inhibit the expression of mannose receptor on macrophages and downstream signaling pathways,then related antibodies production were deduced and immune response was suppressed.lt would inhibit the synovial and bone damage. All these could achieve the goal of anti-rheumatoid arthritis. |
