Study Quality Standard Of Rootlet Coptidis And Advance Quality Standard Of The Crude And Processed Rhizoma Of Coptidis

Rhizoma Coptidis is the dried root of Coptis chinensis Franch., Coptis deltoidea C. Y. Cheng et Hsiao, and Coptis teeta Wall, of ranunculaceae plants. The item of "Rhizoma Coptidis" in the China Pharmacopoeia of2010edition contains "character","identification","check","extractum" and "content determination". It can identify five alkaloids at the same time by TLC, but we found that the spots of Jatrorrhizineand and Columbamine have the same position in the process of the research of quality standard of Rootlet Coptidis; Content determination under QAMS to determine the content of4alkaloids simultaneously, but it is reported that six kinds of alkaloids can be simultaneously determined by HPLC. It will be more conducive to the quality control of the crude and processed rhizoma of Coptidis by improving the content determination method and increasing the types of index components.Rootlet Coptidis is the fibrous root of Coptis chinensis. As a by-product of Coptis, it has not been included in Chinese pharmacopoeia over the years, only standard of traditional Chinese medicine of guangdong province (second volume) has it as the standard for places, which contains the "character","identification","check","extractum" and "content determination"; but the sample preparation and developing solvent were complex and operation is not easy by TLC identification; It is difficult to fully reflect and control the quality of Rootlet Coptidis that Berberine is the only index ingredient of TLC identification and content determination, the objectivity and specificity is poorer.The quality standard of Rootlet Coptidis refer to the quality standard of Rhizoma Coptidis in Chinese pharmacopoeia of2010edition and the standard of traditional Chinese medicine of guangdong province(second volume), and the quality standard has been improved. It is more to the benefit of comprehensive control of quality that the determination of Jatrorrhizine and Columbamine is added to quality standard of Rhizoma Coptidis. The quality standard of Rootlet Coptidis is more comprehensive and specificity than standard of traditional Chinese medicine of guangdong province (second volume).The following work was finished by this standard research:1. Established its character according to descripting characters of Rootlet Coptidis sample and methods of identification by microscopy and TLC.2. According to the appendix IX H, IX K and X A of Chinese pharmacopoeia of2010edition, the moisture, ashe, and enthanol extract of Rootlet Coptidis were determined respectively. 3. External standard method(ESM) and quantitative analysis of multi-components by single marker(QAMS) were used to determine the content of Jatrorrhizine, Columbamine, Epiberberine, Coptisine, Berberine in the crude and processed rhizoma of Coptidis and Rootlet Coptidis; At the same time, the content limit of six alkaloids was established(the content limit of Columbamine was excluded in Rootlet Coptidis)...
