| Objective:On the basis of preliminary research,to study the regulated impact of rats TGF-β1/smads protein signal pathways intervention by Tongmaidasheng-Tablet (TMDST) based on BuShen TiaoChong treating androgen induced sterile rats(ASR).To provide scientific evidence of BuShen TiaoChong on sterile rats and to further study the mechanism of anovulatory infertility.Method:ASR model was established by injecting testosterone propionate subcutaneously to SD female rats of9days age.Then the rats were divided into seven groups:model group,TMDST high-dose group.TMDST medium-dose group,TMDST low-dose group,letrozole group,You gui-pill group and blank control group.The medication was started at80days old until5weeks later,then the blood was taken through femoral artery.The level of serum TGF-β1was detected by enzyme-linked immunosorbent assay,then the bilateral ovaries were stripped quickly,of which the left ovary was froze by liquid nitrogen and the level of Smad4ã€Smad7were detected by immunoblotting assay,the right was fixed in10%methanal and the level of TGF-β1R was detected by immunohistochemistry.Results:1ã€The level of smad7was the highest in model group,compared with other groups except TMDST low-dose group(P<0.01),the difference was significance,but compared with TMDST low-dose group(P>0.05),there was no statistical significance.The level of Smad7was the lowest in TMDST high-dose group and letrozole group.2ã€The level of Smad4was the lowest in model group,compared with blank control groupã€TMDST high-dose group and letrozole group,the difference was significance(P<0.01),and compared with Yougui-pill group,TMDST medium-dose group(P<0.05),but there was no statistical significance with TMDST low-dose group(P>0.05).The level of Smad4was the highest in TMDST high-dose group and Letrozole group. 3ã€The level of TGF-β1was the lowest in model group.Compared with blank control group and letrozole group,the difference was significance(P<0.01),and there was statistical significance with TMDST high-dose group and Yougui-pill group(P<0.05), but there was no statistical significance with TMDST medium-dose group and TMDST low-dose group(P>0.05).The level of TGF-β1was the highest in TMDST high-dose group in all TMDST groups.4ã€Compared with model group,the level of TGF-β1R was improved in other groups among which the positive expression was the highest in TMDST high-dose group and letrozole group,and the positive expression was equal in Yougui-pill group and TMDST medium-dose group.Conclusion:1ã€There was imbalance in the TGF-β/smads protein signal pathways of ASR.TMDST could up regulate the expression of smad4protein and down regulate the expression of smad7protein by regulating the level of smads protein expression to improve the local microenvironment of ovaries.2ã€TMDST could increase the expression of signal transducers TGF-β1. TGF-β1R of ovaries and make the pathway fluent between signal transducers and signal protein,in order to improve ovulation failure.3ã€TMDST based on BuShen TiaoChong could not only regulate the TGF-β1/smads signal pathways of ASR but the local factors of ovaries,so the writer inferred that partial effects was achieved by regulating the ovarian TGF-β1/smads signal pathways. |
