Research On Expression Of Collagen Type â…¢ And PDGF Through Pattern Of Moxibustion And Huaxian â…£ For Bleomycin-Induced Mice Pulmonary Fibrosis

ObjectiveThe purpose of this work was to know the expression of Collagen Type Ⅲ and PDGF in the bleomycin-induced mice with pulmonary fibrosis after taking treatment with moxibustion and huaxian Ⅳ; moreover, we discussed the mechanism of the medicine making effect on pulmonary fibrosis, aimed to find dependable theoretical foundation.Methods130SD rats (200g±20g), sex in half, SPF, were randomly divided into five groups. The first group was untreated(control); the second group was injected with bleomycin; the third huaxian IVwas treated with huaxian IV(traditional Chinese medicine); the fourth group was only used moxibustion; the fifth moxibustion and huaxian IV together were used both methods to treat disease. Each group got30rats, only control group got10rats. The experiment took place in institute of acupuncture and moxibustion Chengdu University of TCM. The Chinese herb was purchased by Chengdu University of TCM affiliated hospital. The huaxian IV contained honghua, sigualuo and so on. Those herbs were decocted by rongle pharmacy. Lavage needle was number16. Except the control group, the other five groups were injected bleomycin in trachea to make rats get pulmonary fibrosis, then immediately upright spinning rats made sure bleomycin was equally diffused. In the meanwhile the animal freely dirked water and ate food. After treatment, at7d,14d,30d, respectively, we killed8rats,took out lung tissue to make HE stain, observed their pathological change, and detected the expression of Ⅲ-C and PDGF.ResultsRT-PCR showed:compared with the control group, expression of Ⅲ-C and PDGF of other four groups were increased (P<0.05); compared with the bleomycin group, the expression of Ⅲ-C and PDGF of the other groups were decreased(P<0.01). In this experiment, revealed the lowest expression of Ⅲ-C and PDGF was moxibustion and huaxian Ⅳ together group. The result stated moxibustion and huaxian Ⅳ together group made better effect than other groups. Western blot explained that compared with the bleomycin group, each group expression of Ⅲ-C had difference.Conclusion1. Moxibustion therapy can relieve pulmonary fibrosis degree of rats by bleomycin induced.2. Huaxian Ⅳ can alleviate pulmonary fibrosis degree of rats by bleomycin induced.3. Therapeutic effect of moxibustion and huaxian Ⅳ together was superior to only using moxibustion or huaxian Ⅳ.4. The mechanism of moxibustion and huaxian Ⅳ together mitigating pulmonary fibrosis degree may through regulating Ⅲ-C and PDGF.
