| BackgroundPsoriasis is a recurrent and chronic inflammatory skin disease. The etiology iscomplicate and it is one of the dermal disease for emphatic study. Through activationon TLR by Imiquimod produced synthesis and secretion of many cytokine andchemotactic factor, and than it leaded to inflammatory reaction and impaired immunefunction. When it is used on mice local skin externally, it will induce Psoriasis-like onmice local skin.Alpha1antitrypsin is a important defensive proteins in physiologicalcirculation, it plays important role in anti-inflammatory, immunoregulation, inhibitoryapoptosis and keeping human physiological circulation,etc.Objectives1.To establish the model of Imiquimod-induced psoriasiform inflammation ofBALB/c mice2.To observe the treatment of AAT on Imiquimod-induced psoriasiform inflammationof BALB/c mice and investigate the related mechanism.Methods1.24female BABL/c mice were randomly divided into four groups: short-terms blankcontrol group(use vaseline externally last seven days), long-terms blank controlgroup(use vaseline externally last fourteen days), shot-terms model control group(use5%Imiquimod last seven days), long-terms model control group(use5%Imiquimodlast fourteen days). Observe general skin lesion and pathological changes.2.60female BABL/c mice were randomly divided into six groups: blank controlgroup, MTX group(positive control group), Model group(negative control group),high-concentration treatment group(3mg/ml AAT), median-concentration treatment group(2mg/ml AAT), low-concentration treatment group(1mg/ml AAT).Observe andcompare each group through general skin lesion (PASI score), pathological changes(BAKER score) and inflammatory cell counts. At last make the statistical analysis.Results1.After the first day it appeared red plaquesã€scale and hypertrophy on model groupmice.The lesions aggravated through the accumulate of Imiquimod.On the eighth daythe symptoms is to extreme,and then it would decreased.2.The high-concentration of AAT have effects on the treatment of psoriasis-like miceskin lesion which induced by Imiquimod(P<0.05),and the effects is better thanMTX(P<0.05).The reducing of inflammatory cell have a positive correlation to theconcentration ofAAT(P<0.05).The median and low concentration of AAT have littleeffect on the treatment(P>0.05).Conclusions1. When using Imiquimod externally on local skin from psoriasis-like, it willsuccessfully obtain Psoriasis-like skin lesion.2ã€AAT treatment on the psoriasis-like mice which induced by Imiquimod is effective.Though reducing the quantity of inflammatory cell to inhibit inflammatoryreaction,but this treatment is independent to theAAT concentration. |
