Objective:In this study,ultraperformance liquid chromatography mass spectrometry(HPLC-MS) was used to profile the metabolites of blood and urine samples fromcolorectal cancer patients and healthy control group.Multivariate statisticalanalysis methods were used to discriminate between the samples and find thebiomarkers of colorectal cancer,besides,inspected the accuracy in distinguishbetween colorectal cancer and health group.It was to provide scientific basisfor search new tumor markers and drug targets of colorectal cancer.Methods:1. Sample preparation:The samples were centrifuged at14000rpm for5min,takethe upper phase for HPLC-MS analysis.It inorer to ensure the integrity ofmetabolic spectrum.2. LC-MS analysis of samples:The blood and urine samples were analyzed byHPLC-TOFMS and obtained metabolic spectrum of the blood or urine from colorectalcancer patients and healthy person.3. Data analysis:Multivariate statistical analysis was used to analysis thedata and check the difference between colorectal cancer patients and healthyperson.It was to find out the metabolites that distinguish between experimentalgroup and control group. At the same time,inspected the diagnostic accuracy ofthese metabolites for colorectal cancer.Results:1. In the research of blood samples,the clear separation of two groups wasobserved in the PCA scores plot where each point represents a sample.Bystatistical analysis,18important discrepant metabolites were found in bothpositive and negative ion mode.Finally,we finded6key discrepant metabolites as biomarkers of colorectal cancer by the ROC curve and mean histogram.And itverified them have a high accuracy and specificity in distinguish betweencolorectal cancer and health group.2. Similarly,in the research of urine samples,the acquired metabonomic datawas analyzed by PCA. From the PCA scores plot,we can see two groups were clearlydistinguished from each other.We also found19important discrepant metabolites.Finally,we finded8key discrepant metabolites as biomarkers of colorectal cancerby the ROC curve and mean histogram.And it verified them have a high accuracyand specificity in distinguish between colorectal cancer and health group.Conclusion:In this study,the HPLC-MS method was used for the metabonomic analysis ofblood and urine samples from colorectal cancer patients and healthy person,andobtained the metabolic spectrum.Then we finded the biomarkers of colorectalcancer by multivariate statistical analysis methods and verified them have havea high accuracy and specificity in distinguish between colorectal cancer andhealth group. The results of this study demonstrated that the metabonomics isa powerful and useful tool for the analysis of the cancer.
