Objective:To observe the effect of hepatic and renal functions with the treatment of singlelarge dose of simvastatin and small doses of simvastatin combined with ezetimibe inrats arteriosclerosis model,and the he impact of level of immune factors in serumduring the process of atherosclerosis.Method:446months old healthy Wistar male rats are divided randomly into five groups:blank group、model group、low dose of simvastatin (10mg/kg·d) treatment group、thehigher dose of simvastatin (20mg/kg·d)treatment group and low dose of simvastatin(10mg/kg·d) combine with ezetimibe(5mg/kg·d) treatment group.Except the blankgroup,the rats in the other groups are intraperitoneal injection with vitamin D3for thefirst3days,and feed with high-fat diet continuously for8weeks to made into ASmodel.Then the corresponding dose of drug are given through irrigation stomach ineach group for4weeks.Measure the level of TG、TC、LDL-C、HDL-C、ALT、AST、Scr、BUN、UA、U-pro、Ucr、CCr and IFN-r、IL-4in serum before and after the drugtreatment. Observe the change of thoracic aortic pathology and renal tissues usingHE-staining by light-microscope.Results:1.The model group(C group) contrast with the blank group(N group) have ahigher level of TC、LDL-C、TG,and lower level of HDL-C,compared the two groupsare statistically different;the level of TC、 LDL-C、TG are decreased in the drugtreatment group,the SE group decreased most among them.2、The rats in the C group have higher level of ALT、AST compare with the N group,and the difference is statistically significant;the level of ALT、AST are decreasedin the drug treatment group,the SE group decreased most among them.3.The C group contrast with the N group have a higher level of BUN、UA、U-pro,and lower level of Ccr,compared the two groups are statistically different;thelevel of BUN、UA、U-pro are decreased and Ccr is increased in the drug treatmentgroup,the SH group changed most among them.Glomerular volume of C group arebigger than N group,and mesangial cell hyperplasia slightly,mesangial matrixincreased; glomerular volume of drug treatment group are smaller than Cgroup,reduced hyperplasia,the lesions of SH group are the mildest.4、The serum concentration of IFN-r increased significantly in C group comparedwith N group,and the treatment of high dose of simvastatin is the most efficient indecreasing the level of IFN-r than S group and SE group,and the level of IFN-r isrelate to the level of lipid,and there is a positive correlation between the level of IFN-rand the the level of TC、TG and LDL-C,there exists negative correlation between thelevel of IFN-r and the level of HDL-C;the level of IL-4have no difference between Cgroup and N group,and have no changed after drug treatment.Conclusions:1、Low doses of simvastatin joint ezetimibe can significantly reduce the level oflipid,and improve the liver function induced by hyperlipemia.2、 The higher dose of simvastatin can better protect the kidney function,andeffectively improve the lesion of glomerular.3、The higher dose of simvastatin can better reduce the level of IFN-r,to improvethe immuno-inflammatory response in the process of atherosclerosis.The impact oflevel of IL-4is not significant during the process of atherosclerosis.
