Objective:To establish a common obstructive jaundice SD rats by surgical ligation Sprague-Dawlwy(SD)rats, sbile duct, the prelimnary study on the impact of selenium when bilirubin and hepatic cell morphology changes in rats with obstructive jaundice.Methods:60 healthy adult male SD rats were used as experimental subjects, body weight250±15g,wrer randomly divided into three groups of 20:groupA(control group):virtual surgery group; group B(control group):bile duct ligation+free drinking water feeding:groupC(experimental group):bile ligation+selenium fed group.Group A: After successfully anesthetized rat peritoneal conventional open not only to separate the common bile duct ligation,and then the abdomen was closed layer by layer,normal drinking water feeding;group B After the success of anesthesia conventional open common bile duct neat the duodenum isolated rat peritoneal double direction ligation of the common bile duct,and then the abdomen was closed,normal drinking water feeding;Group C:After the success of anesthesia conventional open isolated rat peritoneal bile ductligation double direction close to the duodenum,and then the abdomen was closed,on the first day after the start of infusion Se Kang (Anhui Province Huaxin sheng Pharmaceutical Co.Ltd).daily 10mg/Kg and feed.In the first two days after surgery,four days,six days,eight days, ten days five times in three groups of rats were randomly selected four rats,after the success of anesthesia laparotomy,abdominal aortic blood,test to compare the serum levels of total bilirubin(TB),direct bilirubin(DB),while the liver tissue with HE staining of liver tissue pathological changes.The data obtained using the SPSS 17.0 statistical software for data analysis, among the three groups were compared using ANOVA,P<0.05 was considered statistically significant.The experimental data as meant± standard deviation(x±s) described below.Significant test test with t.The result1.successfully established animal model of obstructive jaundice in rats.2.1n terms of the three groups of rats bilirubin between group B and group A statis statistically significant difference(P<0.05),the difference between group C and A groups was statistically significant(P<0.05),group C and B compared to serum at each time point TB and DB were significantly increased,and the trend slowed down compared with group B(P<0.05)3.HE visible staining of liver tissue morphology,A group of normal lobules and portal structure,no inflammatory cell infiltration,no bil duct hyperphlasia. Group B visible,appear after two days surrounding liver tissue lobules several periportal in-flammatory cell infiltration,periportal biliary epithelial thickening visible;after four days seen in liver tissue lobules and portal area a significant increase in inflammatory cell,biliary epithelial cells than before thickening and a small anount of points,flake necrosis;gradually increased after liver injury,necrosis until eight days after the first appearance of liver tissue is gradually increasing.Group C visible,appear after two days surrounding liver tissue lobules several periportal in flammatory cell infiltration, periportal biliary epithelial thickening visible.After the fourth day to 10 days after surgery compared with group B liver lobule and periportal tissue inflammatory cell infiltration lightet,bile duct hyperplasia relatively mild,relatively small range of necrosis.In conclusionLrat model of obstructive jaundice causde by bile duct ligation in rats in the early postoperative period in group B of rat liver edges of Dayton,the surface scattered bilirubin deposition surface of the liver with prolonged obstruction appear flaky bilirubin deposition,pathlolgy see periportal inflammatory cell infiltration,bile duct hyperplasia,liver tissue punctuate,shape and focal necrosis.Bilirubin gradually increased after the first eight days to reach the highest value.2.C group on the frist day after the start of the rats fed Se Kang,Group C rat liver early appearance and no significant difference between group B,with the continued obstruction of the time it is deposited in liver surface bilirubin than B group light, pathological,liver punctate necrosis, necrosis of small,elevated levels of serum bilirubin trends and decreased compared to group,this shows that when rats with obstructive jaundice selenium can increase serum bilirubin slowing trend.3.Selenium liver injury has a protective effect.
