Objective:Explore the expression of plasma miR-21, miR-210 in pancreatic cancer and its relationship with benign and malignant. To investigate the difference of miR-21 and miR-210 in plasma and tissue of pancreatic cancer. some tissue have got by EUS-FNA,discussion EUS-FNA for the diagnosis of early pancreatic lesions and pancreatic tumors. Explore the diagnostic value of miRNA and serum CA199, CA242, CEA of pancreatic cancer.Methods:Plasma samples from sixty patients with pancreatic cancer(PCa), twenty patients with chronic pancreatitis(CP) and ten normal people.Ten tissue samples from pancreatic cancer. Total RNA was extracted from plasma samples and tissue samples for measurement of miR-21、miR-210 levels using real-time PCR. Simultaneously, serum tumor markers carbohydrate antigen 199(CA199), carbohydrate antigen 242 (CA242) and carcino-embryonic antigen(CEA) were determined. Analysis the relationship of plasma miR-21, miR-210 levels between PCa group and CP group, normal group. The relationship between plasma miR-21, miR-210 levels and CA199,CA242, CEA, tumor diameter of pancreatic cancer patients was analyzed. The differential of miR-21、miR-210 levels between plasma and tissue in patients with PCa was analyzed. The diagnostic efficiency from plasma miRNAs and CA199,CA242,CEA in PCa patients was evaluated.Results:1. The relative abundance of plasma miR-21、miR-210 were significantly higher in the PCa group than in the CP and normal group (3.96±3.74 vs 1.11±2.76,0.145±2.68; 4.115±4.51 vs 1.49±3.94, -1.73±4.82; 均 P<0.01).2. The relative abundance of miR-21、miR-210 in PCa tissue was higher than plasma(5.28±7.87 vs 3.89±6.55,4.69±2.54 vs 3.25±1.98, 均 P>0.05)。3. Plasma miR-21 levels was negatively correlated with CA199 in PCa patients, Plasma miR-210 levels showed no significant correlation with CA199 or CA242 or CEA or tumor diameter in PCa patients.4. The AUC-ROC in the PCa group vs normal group, PCa group vs CP group, PCa group vs CP group and normal group of plasma miR-21,miR-210 were 0.817,0.870. 0.752,0.788.0.773,0.816.respectively.5. Analyzing by a binary logistic regression model find that: the sensitivity and specificity in the PCa group vs normal group, PCa group vs CP group, PCa group vs CP group and normal group by Combination of plasma miR-21, miR-210 and serum tumor markers CA199, CA242,CEA can be improved.Conclusion:1. MiR-21, miR-210 were highly expressed in PCa blood. And can distinguish between cp and normal people.2.Plasma miR-21, miR-210 has no relationship with the maximum diameter of PCa tumors.3. MiR-21, miR-210 were highly expressed in PCa tissue and blood, The expression of miR-21, miR-210 in PCa tissue and plasma has no differences.4.Combination of plasma miR-21, miR-210 and serum tumor markers CA199, CA242,CEA can improve the diagnostic value in PCa.
