Effect Of Deep Hypothermic Circulatory Arrest On Cytoskeletal Proteins Of MPTP In Hippocampus Neuron Of Rats | | Posted on:2016-11-28 | Degree:Master | Type:Thesis | | Country:China | Candidate:Y Q Deng | Full Text:PDF | | GTID:2284330470467217 | Subject:Surgery | | Abstract/Summary: | PDF Full Text Request | | [Objective] To study the effect of deep hypothermic circulatory arrest (DHCA) oncytoskeletal proteins of mPTP in Hippocampus neuron of rats and mechanism of hypothermia in brain protection through inhibiting mitochondrial apoptosis.[Methods] Thirty-five SD rats were divided randomly into three groups:Deep hypothermic circulatory arrest (DHCA) group (n=10), Normal temperature circulatory arrest (NTCA) group (n=10) and Normal control (NC) group (n = 10), the next five rats were usedas the blood donor for other rats. The animal model of closed-chest DHCA was established in rats by means ofexternal cannulation. In DHCA group, ten minute’s circulatory arrest was performed after the temperature of rectum between 20℃-24℃ (by external circulation and body surface cooling); in NTCA group, ten minute’s circulatory arrest was performed in normal temperature by the same method; in NC group, ten minute’s external circulation without circulatory arrest. Hippocampus were dissociated after external circulation and then VDAã€ANTand CYPD was measured by WB and RT-PCR, the results were analyzed by software SPSS 17.0. Data were expressed as the mean± standard error of the mean. Differences among groups were compared by one-way analysis of variance with post hoc comparisons performed using the Student-Newman-Keuls test of multiple comparisons. A difference of p<0.05 was considered significant.[Results] The levels of VDACã€ANT land CYPD expression of NTCA group and DHCA group were much higher than NC group (P<0.05). The levels of VDAC, ANT land CYPD expression of NTCA group were much higher than DHCA group (P<0.05)[Conclusions] Ischemia can induce the expression of VDACã€ANT and CYPD while deep hypothermia can reduce the expression apparently.
